AAA+ proteases, ClpX, ClpP, ClpXP, function, structure, bacteria, virulence, physiologyIntracelulární proteolýza je základní regulační proces ovlivňující buněčnou fyziologii. Jelikož proteolýza nevratně ničí proteiny, je nutné, aby byl tento proces přísně kontrolován. U bakterií jsou základními kameny regulované proteolýzy AAA+ proteiny. Tyto proteiny se skládají ze dvou funkčních domén a to AAA+ šaperonové domény a proteázové domény. Jednou skupinou těchto AAA+ proteinů je rodina Clp proteinů, u které jsou funkční domény tvořeny samostatnými proteiny. Jedním z těchto proteinům je hexamerická AAA+ unfoldáza ClpX. Tato unfoldáza může interagovat s vysoce konzervovanou proteázou ClpP za tvorby proteolytického komplexu ClpXP. Tento proteolytický komplex využívá energii vazby ATP a jeho hydrolýzu k rozbalení a translokaci specificky značeného substrátu do degradační komory ClpP. Rozpoznání substrátu je zprostředkováno vazbou ClpX na krátké nestrukturované sekvence nazývané degradační značky. ClpX rozpoznává velké množství degradačních značek, avšak nejzásadnější je rozpoznání ssrA degradační značky, která je výstupem tmRNA systému záchrany ribozomu. I když ClpX ve spojení ClpP ovlivňují celou řadu buněčných pochodů, jako je exprese virulentních faktorů či adaptace na stresové faktory, ClpX zastává i role zcela nezávislé na ClpP. Intracellular proteolysis is an essential regulatory process that affects cellular physiology. Since proteolysis destroys proteins irreversibly, this process must be strictly controlled. The AAA+ proteins are the key factors in regulated proteolysis in bacteria. These proteins consist of two functional domains, the AAA+ chaperone domain and the protease domain. One particular group of these AAA+ protein is the Clp protein family. Functional domains of the Clp family are formed by seperate proteins. The hexameric unfoldase ClpX is a member of this protein family. This unfoldase can interact with the highly conserved ClpP protease to form a ClpXP proteolytic complex. This proteolytic complex utilizes the energy of ATP binding and hydrolysis to unfold and translocate the specifically tagged substrate into the ClpP degradation chamber. Substrate recognition is mediated by the binding of ClpX to short unstructured sequences called degradation tags. ClpX recognizes several degradation tags, but the most important one is recognition of the ssrA degradation tag, which is the output of the tmRNA ribosome rescue system.
