Bombus lucorum, Bombus terrestris, reductase fatty acidsV období páření čmeláků na své letové trase značkuje samec různé objekty druhově specifickým samčím značkovacím feromonem k nalákání samičky. Na syntéze feromonů, která probíhá v labiální žláze čmeláčích samců, se podílejí enzymy zvané reduktasy mastných kyselin (dále jen reduktasy). Katalyzují redukci mastných kyselin vázaných ve formě acyl-koenzymu A na jejich alkoholy. V této práci jsem funkčně charakterizovala dvě reduktasy vysoce exprimované v labiální žláze s cílem ověřit jejich úlohu v biosyntéze druhově specifických feromonů, u dvou nejběžnějších druhů českých čmeláku Bombus lucorum (čmelák hájový) a Bombus terrestris (čmelák zemní). Exprimovala jsem zmíněné reduktasy v kvasinkách Sacccharomyces cerevisiae. Potvrdila jsem heterologní expresi reduktas a připravila lipidové extrakty z kvasinkových buněk, ve kterých byly pomocí metody hmotnostní spektrometrie ve spojení s plynovou chromatografií identifikovány alkoholy odvozené od mastných kyselin s velmi dlouhým řetězcem. Nejvíce zastoupeným analyzovaným alkoholem extraktu z kvasinek, které heterologně exprimovaly čmeláčí reduktasy, byl oktadekanol, ikosanol a dokosanol. Tyto výsledky nás vedou k závěru, že enzymy reduktasy se podílí na biosyntéze alkoholů s dlouhými uhlovodíkovými řetězci, které jsou složkami značkovacích feromonů...
In the mating season, bumblebee males leave pheromone track on different objects on their flight routes to attract females of the same species. Synthesis of pheromones, which takes place in the labial gland of bumblebee males, involves enzymes called fatty acid reductases (hereinafter reductases). They catalyse the reduction of fatty acids bound in the form of acyl-coenzyme A to the alcohols. In this work, I functionally characterized two reductases highly expressed in labial gland of the two most common species of bumblebees Bombus lucorum and Bombus terrestris in order to verify their role in the biosynthesis of species specific male pheromones. I expressed these reductases in yeast Sacccharomyces cerevisiae. I confirmed the heterologous expression of reductases and prepared lipid extracts from yeast cells, in which we identified by gas chromatography with mass spectrometric detection (GC/MS) alcohols derived from fatty acids with long chain. These were octadecanol, icosanol and docosanol.These results lead us to conclusion that reductases are involved in the biosynthesis of alcohols with long hydrocarbon chains, which are components of marking pheromones of mentioned bumblebee species.
