Torymidae, Toryminae, Microdontomerini, molecular characters, morphological characters, phylogenyNadčeleď chalcidky (Chalcidoidea) je druhově nejbohatší skupinou řádu blanokřídlého hmyzu (Hymenoptera) a v současnosti zahrnuje celkem 22 čeledí. Jednou z čeledí je také čeleď krásenkovitých (Torymidae), jejíž monofyletičnost je v současnosti intenzivně diskutována. Tato diplomová práce se zaměřuje na tribus Microdontomerini (Torymidae: Toryminae). Druhy tohoto tribu jsou většinou drobné a nenápadné (délka těla 2 až 4 milimetry). Současný pokrok ve fylogenezi chalcidek umožňuje vytvořit robustní fylogenetickou hypotézu tribu Microdontomerini jako součást výzkumu fylogeneze a klasifikace chalcidek. Můj přístup kombinuje molekulární a morfologické metody tak, aby byl výsledný fylogram co nejvěrohodnější. Pro analýzu distribuce morfologických znaků bylo vybráno 33 zástupců reprezentujících všechny rody tribu Microdontomerini a 10 vnějších taxonů. Pomocí skenovacího elektronového mikroskopu Hitachi S-3700N-VP jsem u těchto vybraných taxonů zkoumal 33 morfologických znaků. Pro 117 taxonů byla získána sekvenční data z 5 jaderných (18S, 28SD2, 28SD3-5, EF1alfa, Wingless) a 3 mitochondriálních (COI-LCOHCO, COI-JerryPat, CytB) genových úseků, což ve výsledku vytvořilo dataset o 387 Kbp. Superfamily Chalcidoidea (chalcid wasps) represent the most species-rich group of the Hymenoptera and includes 22 families. Family Torymidae is one of those families and it's monophyly is currently intensively discussed. This particular thesis aims at the tribe Microdontomerini (Torimidae: Toryminae). Species of this tribe are usually minute inconspicuous wasps (body length ranges from 2 to 4 mm). Recent progress in phylogenetics of chalcid wasps allowes us to formulate robust hypothesis of Microdontomerini evolution, as a component of broader study of Chalcidoidea phylogeny and classification. My approach combines molecular and morphological evidence to formulate the most plausible evolutionary scenario. Dataset of 35 mostly morphological characters for 33 taxa including all existing Microdontomerini genera and 10 outgroups has been scored. Almost all characters has been studied and documented using Hitachi S-3700N-VP scanning electron microscope. In 117 taxa, 5 nuclear genes segments (18S, 28SD2, 28SD3-5, EF1alfa, Wingless) and 3 mitochondrial genes segments (COI-LCOHCO, COI-JerryPat, CytB) were sequenced, to get together the final molecular dataset of 387 Kbp.
