Integrator complex, INTS11, RNA Polymerase II, Transcription, Neurodevelopmental disordersIntegrator komplex je proteinový komplex, specifický pro mnohobuněčné organismy, se dvěma enzymatickými aktivitami; endonukleázovou a fosfatázovou. Tento komplex tvoří 15 podjednotek, INTS1-INTS15, které se seskupují do modulů. Integrator byl původně označen jako komplex zapojený do procesování 3'konců UsnRNA, avšak později bylo popsáno, že působí i na jiných nekódujících RNA substrátech. Nejen to, data ukázala, že se dále účastní celkové regulace transkripce kódujících genů, při které ovlivňuje pausování RNA Polymerázy II v promotor-proximálních oblastech, elongaci i terminaci transkripce. Některé podjednotky Integratoru byly popsány také v odpovědi na poškození DNA. Mutace v pěti podjednotkách Integratoru jsou spojeny se závažnými neurologickými vadami a zároveň deregulovaná exprese podjenotek Integratoru byla propojena se vznikem rakoviny. Přestože mnoho funkcí Integratoru bylo objeveno a jeho struktura byla z velké části popsána, rozsáhlejší výzkum je potřeba k objevení nových regulačních mechanismů Integratorových podjednotek a deregulaci s nimi spojenou, jež může dále vést s k onemocněním spojenými s Integratorem. Integrator complex is a metazoan-specific protein complex with dual enzymatic activity; endonuclease and phosphatase. The 15 subunits, INTS1-INTS15, form this complex and are arranged into modules. Integrator was originally identified as a complex involved in the 3'end processing of UsnRNAs, and later also described to act on other non-coding RNA substrates. Not only that, but evidence also sheds light on its function in overall transcriptional regulation of protein-coding genes, further influencing promoter-proximal pausing of RNA polymerase II, transcription elongation, and transcription termination. Some Integrator subunits have also been reported to participate in the DNA damage response. Interestingly, mutations in five Integrator subunits are linked to severe neurological diseases, and deregulated expression of Integrator has been linked to cancer development. Even though several functions of Integrator have already emerged and the structure of the complex has been largely described, extensive research is still needed to uncover novel regulatory mechanisms involving Integrator subunits, the deregulation of which may further contribute to the pathology of Integrator- related diseases.
