Azurin je jedním z nejpoužívanějších modelových systémů pro studium oxidačně-redukčních a elektron transportních procesů v proteinech. V této práci jsme použili metodiku kovalentního fotochemického zesítění (PIXL) pro studium oligomerizace azurinu v roztoku. Zároveň jsme studovali vliv L-2- amino-5,5-azi-hexanové kyseliny (foto-Met), strukturního světlem indukovatelného analogu přirozené aminokyseliny Met, na elektron transportní procesy v azurinu. Byla provedena optimalizace podmínek exprese rekombinantního azurinu v auxotrofních buňkách E. coli B834 za účelem získání azurinu s maximální mírou inkorporace foto-Met (70% inkorporace určená MALDI-TOF hmotnostní spektrometrií). Současně modifikací purifikačního protokolu (např. rozbití buněk, kyselá precipitace proteinů, zavedení kovového ligandu při sonikaci buněk) jsme zvýšili čistotu a výtěžek preparátu a zkrátili dobu purifikace. Získané preparáty azurinu (přirozený protein (WT) s Met, WT s foto-Met a "All-Phe" mutant (všechny Trp/Tyr zaměněny za Phe) s foto-Met) byly po ozáření UV světlem (metodika PIXL) charakterizovány pomocí UV-VIS spektroskopie a SDS-PAGE. Electron transport processes are an extremely important field of study in modern biochemistry and structural/functional proteomics. Azurin is one of the most utilised model systems for study of redox and electron transport processes in proteins. We have utilised photo-induced crosslinking (PIXL) to study oligomerization of azurin in solution and the effect of L-2-amino-5,5-azi-hexanoic acid (photo-Met) - a structural photoinducible analogue of canonical amino acid Met - on electron transport processes in azurin. The optimisation of expression conditions of recombinant azurin in auxotrophic E. coli B834 cells was done to maximise photo-Met incorporation percentage in azurin sequence (70% incorporation was measured via MALDI-TOF mass spectrometry). Through the optimisation of purification protocol (example: cell disintegration, acid precipitation of proteins, adding metallic ligand during cell sonication) we have increased the purity and yield of final product and reduced the purification time.
