7 1. Úvod Fenomén restrikce a modifikace byl poprvé popsán v 50. letech minulého století. Studium restrikčních enzymů umožňujících manipulaci s molekulami DNA a konstrukci rekombinantních molekul dalo vznik novému směru molekulární biologie - genovému inženýrství. Právě intenzivní studium restrikčních enzymů označovaných dnes jako Typ I umožnilo objev restrikčních endonukleas, které jsou označované jako Typ II a v současné době běžně používané v praxi. V nedávné době došlo v souvislosti s postupujícím sekvenováním bakteriálních genomů, které odhalilo široké rozšíření těchto restrikčních enzymů mezi mikroorganismy v přírodě, k oživení zájmu o restrikční enzymy Typu I. Restrikčně modifikační (R-M) enzymy mají důležitou funkci při ochraně bakteriálních buněk před vstupem cizorodé DNA v procesech konjugace, transfekce a transformace. Proto má studium těchto enzymů i značný praktický význam pro techniky genového inženýrství, neboť efektivnost těchto procesů může být výrazně snížena, jestliže použitá DNA není vhodně modifikována. Tato vlastnost vedoucí k zabránění vstupu cizí nemodifikované DNA do buňky může být naopak využita v boji s fágovou lyzí kmenů producentů. Dále se tyto enzymy zřejmě účastní široké škály molekulárních procesů probíhajících v buňce jako je replikace, genetická rekombinace a reparace DNA....
