Metamphetamin, striatum, prefrontal cortex, hippocampus, prenatal exposition, microarrays, real-time PCRÚvod: Metamfetamin je droga často zneužívaná těhotnými ženami závislými na drogách a je současně jednou z nejčastěji užívaných drog v České republice. Tato droga prochází snadno placentární bariérou do organismu plodu a může negativně ovlivňovat nejen matku, ale i prenatální vývoj plodu. Cíle:V rámcigrantového projektu GAČR: 14-03708S byly zjišťovány dlouhodobé účinky prenatální expozice metamfetaminu. Bylo zkoumáno, zda prenatální expozice metamfetaminem ovlivňuje generaci potomků exponovaných samic na úrovni exprese genů ve specifických oblastech mozku, a to ve striatu, hipokampu a prefrontálním kortexu. Metody: Potkanům byly odebírany vybrané části mozku a v těch byly metodou microarray hybridizace a real-time PCR stanoveny změny exprese vybraných genů. Výsledky: V rámci statistického vyhodnocení microarray hybridizace nevyšla změněná exprese u žádného z testovaných genů signifikantně. Byly naměřeny pouze hraniční hodnoty pro 13 genů, které byly dále testovány metodou real-time PCR. Po statistickém vyhodnocení real-time PCR byla signifikantně změněná exprese nalezena u 2 genů vztaženým ku jedné ze dvou endogenních kontrol. Signifikantně zvýšená genová exprese u genu DRD3 a TACR3 byla nalezena ve striatu, v dalších částech mozku změna exprese nalezena nebyla. U genu FOXP2 byly naměřeny hraniční...
Introduction: Methamphetamine is a drug frequently abused by drug-addicted pregnant women and also one of the mostcommonly used drugs in the CzechRepublic. This drug passes easily through a placental barrier into the fetus. Thus it can negatively affect not only the mother but also the prenatal development of her offspring. Objectives: In the framework of the grant project GA CR: 14-03708S, the long-term effects of prenatal exposure to methamphetamine were detected. It was determined whether the prenatal methamphetamine exposure affects the generation of offspring of exposed females at the level of gene expression of genes in specific regions of the brain, striatum, hippocampus and prefrontal cortex. Methods: In the selected parts of the brain, which were removed from the rat, the microarray hybridization and the real-time PCR to express changes in expression of selected genes were performed. Results: Statistical analysis of microarray hybridization did not show the significantly altered gene expression in tested genes significantly. Only boundary values for 13 genes were measured, which were further tested by real-time PCR.After a statistic evaluation of real-time PCR, the significantly altered expression was found in 2 genes.
