Neovaskularizace je nezbytnou součástí fyziologického vývoje živých mnohobuněčných organizmů. Umožnuje jedinci růst a vyvíjet se tím, že zprostředkovává tkáním přísun živin a kyslíku. Ze stejného důvodu je nedílnou součástí progrese nádorů, které ke svému růstu také potřebují živiny. Proto je v současné době neoplasmatická neovaskularizace zkoumána jako jeden z možných cílů protinádorové terapie. Nádor s jednou z nejvyšších úrovní vaskularizace je glioblastom multiforme, nejčastější primární nádor mozku. Cílem bakalářské práce je v první řadě shrnou principy fyziologické neovaskularizace se zaměřením na proces angiogeneze a poskytnou přehled faktorů, které je ovlivňují. V další části práce je pozornost kladena na popis specifických patologických mechanizmů neovaskularizace v glioblastomu multiforme a je poskytnut stručný přehled aktuálních možností antiangiogenní terapie.
Neovascularization processes are crucial components for proper develop of multicellular organisms. They are necessary part of body growth. Because of them, body tissues are suply by nutrition and oxygen. Unfortunately in the same way like in the healthy body these processes also help in tumor growth and progression.That is the reason why neoplazmatic neovascularization is being investigated as a possible target of antitumor therapies. One of the greatest level of tumor vacularization is asociated with glioblastoma, the most common primary tumor of human brain. There are a severel aims of this thesis. First of all, discuss mechanisms of neovascularizatin under physiological conditions and introduce factors which are involved in this complex proces. Second part of this thesis payd attention on specific glioblastoma nevascularization mechanisms and provides a brief overview of possible antiangiogenesis therapeutic approaches.
