FFPE, formaldehyde, quality, DNA, fixationFormaldehyd je široce používané fixační činidlo. Jeho výhodou je nízká cena, jednoduché použití a výborné fixační vlastnosti, především rychlé pronikání do tkání, dobré zachovávání morfologických struktur tkáně a kompatibilita s následnými histologickými analýzami. Jeho nevýhodou jsou však negativní účinky na nukleové kyseliny. Vlivem jeho působení dochází k modifikaci primární struktury deoxyribonukleové kyseliny (DNA, z angl. deoxyribonucleic acid), její fragmentaci a vzniku vazeb na proteiny, které znesnadňují izolaci takovéto DNA. Míra negativních účinků formaldehydu je závislá na mnoha faktorech. V této práci byl zkoumán vliv chemického složení roztoku formaldehydu (ředění roztoku, přítomnost pufru či kyseliny mravenčí) a vliv délky fixace. Posuzováno bylo množství DNA a míra fragmentace DNA získané z fixovaných tkání, a to metodou kvantitativní polymerázové řetězové reakce, fluorescenčním stanovením koncentrace DNA a pomocí čipové elektroforézy na přístroji bioanalyzér Agilent 2100. Kvalita a kvantita získané DNA byla následně otestována také při stanovení DNA profilu pro identifikační účely analýzou STR (tandemová opakování krátkých motivů; z angl. short tandem repeats). Formaldehyde is widely used fixative. Its advantages are low cost, simplicity of use and good fixation traits, which are fast tissue penetration, good preservation of morphological structures and compatibility with downstream histological applications. Formaldehyde disadvantages are negative effects on nucleic acids. Formaldehyde solutions modify primary structure of deoxyribonucleic acid (DNA), fragment DNA and create protein- DNA covalent bonds that hinder DNA isolation procedures. Level of negative effects of formaldehyde is dependent on many factors. Effect of formaldehyde chemical composition (formaldehyde dilution, presence of buffer or formic acid) and effect of fixation length were studied in this work. On DNA extracted from fixed tissues, DNA quantity and level of DNA fragmentation were studied by quantitative polymerase chain reaction, fluorescence assay for DNA quantification and by on-chip electrophoresis on bioanalyzer Agilent 2100. Quality and quantity of acquired DNA were tested by DNA profile determination for identification purposes using STR (short tandem repeats) analysis.
