NK cells, NKp80, AICL, recombinant expression, refoldingPřirozené zabíječské buňky neboli NK buňky jsou nedílnou součástí přirozené imunity a plní funkci rozpoznání a usmrcení nádorových a virem infikovaných buněk. Jejich funkce je regulována signály produkovanými díky interakci inhibičních a stimulačních receptorů na svém povrchu se svými specifickými ligandy na povrchu cílové buňky. Aktivačním receptorem je také NK receptor NKp80, který tvoří velmi specifický komplex s buněčným receptorem AICL, přičemž oba tyto receptory spadají do rodiny receptorů podobným lektinům C-typu. Zvýšená exprese receptoru AICL je přednostně specifická pro nádorové buňky myeloidního charakteru. Tato diplomová práce popisuje produkci mutované formy AICL expresí v bakteriích Escherichia coli BL21 Gold (DE3) a následnou izolaci a in vitro renaturaci cílového proteinu. V předchozí studii bylo zjištěno, že lichý počet cysteinů v extracelulární lektinové doméně AICL způsobuje špatné sbalení proteinu. Záměnou lichého cysteinu za serin v poloze 87 bylo dosaženo zisku stabilní rozpustné formy AICL se sudým počtem cysteinů v konzervovaných pozicích, charakteristickým pro receptory C-lektinového typu. Správnost tvorby disulfidových můstků mezi cysteiny byla ověřena hmotnostní spektrometrií. Zisk značného množství proteinu umožnil nasazení krystalizačních pokusů v širokém spektru...
Natural killer cells, or NK cells are an integral component of innate immunity and fullfills the function of recognizing and killing tumor and virus-infected cells. Their function is regulated by signals produced by the interaction of inhibitory and stimulatory receptors on their surface with their specific ligands on the targer cell surface. NKp80 is an activating receptor of NK cells and forms specific complex with cell receptor AICL, both of which belong to the family of C-type lectin-like receptors. Overexpression of AICL receptor is preferably specific for tumor cells of myeloid character. This master's thesis describes the production of AICL mutated form by expression in Escherichia coli BL21 Gold (DE3) followed by isolation and in vitro renaturation of the target protein. In a previous study it was found that an odd number of cysteines in the extracelular lectin domain of AICL causes wrong folding of the protein. Substituting an odd cystein for serine at position 87 lead to stable soluble form of AICL with an even number of cysteines in conserved positions, typical for CTLD receptors. Correctness of the formation of disulfide bonds between cysteines was verified by mass spectrometry.
