Cognitive flexibility, dynamic environment, schizophrenia, rat, Carousel maze, dizocipline, spatial orientation4 Abstrakt Kognitivní flexibilita je schopnost přizpůsobovat své jednání dynamicky proměnlivému okolnímu prostředí. Společně s procesy pracovní paměti, pozornosti, motivace a dalšími je řazena mezi exekutivní funkce. Je důležitá v procesu změny kontingence podnět-odpověď, v okamžiku, kdy již původní podnět dále nevede k odměně. Podle povahy změny lze rozlišit tři druhy kognitivní flexibility: přeučování (reversal), intradimenzionální set shifting a extradimenzionální set shifting. Procesy kognitivní flexibility jsou zpracovávány v fronto- striato-thalamickém okruhu s četnými spojeními s jinými mozkovými oblastmi, zejména s limbickým systémem. Deficit kognitivní flexibility patří mezi kognitivní příznaky schizofrenie, jejíž potkaní animální model byl využitý v experimentální části práce. Cílem práce bylo cílem otestovat deficit kognitivní flexibility u potkaního modelu schizofrenii podobného chování v Kolotočovém bludišti ve čtyřech variantách úlohy s různou kognitivní zátěží. Animální model schizofrenie byl vyvolaný akutním podáním nonkompetitivního antagonisty NMDA receptorů MK-801 v dávkách 0,05 a 0,10 mg/kg intraperitoneálně. Přeučování v této úloze po podání těchto dávek MK-801 poškozeno není, ale byl pozorován signifikantní deficit v procesu extradimenzionálního set shiftu u skupiny s dávkou 0,10 mg/kg...
5 Abstract Cognitive flexibility is an ability to adapt a behavior according to a dynamic changes in an environment. It belongs to executive functions, along with the processes of working memory, attention, motivation and others. Flexibility is important in the process of changing "stimulus-response" contingency at the time when the initial stimulus no longer leads to reward. According to the nature of the changes we can distinguish three types of cognitive flexibility: reversal learning, intradimensional set shifting and extradimensional set shifting. The processes of cognitive flexibility take place in fronto-striato-thalamic circuit with numerous connections to other brain regions, especially the limbic system. Deficit in cognitive flexibility belongs among cognitive symptoms of schizophrenia, a psychiatric illness we studied in the experimental part of this work using its analogue in rat. The aim of the experimental work was to test cognitive flexibility deficit in the rat model of schizophrenia-like behavior in the Carousel maze in four versions of task with different cognitive load.
