Vertikálně diferencovanou centrální část kolonie tvoří dvě morfologicky i fyziologicky odlišné populace buněk, které jsou výrazně oddělené. V horní části kolonie se vyskytují takzvané U buňky, v dolních částech se nachází takzvané L buňky. Molekulární mechanismy vertikální diferenciace jsou z velké části neznámé. Hlavním cílem této práce bylo analyzovat, zda a jak změněná hladina vybraných proteinů signálních drah monitorujících dostupnost živin ovlivňuje vývoj a diferenciaci kvasinkových kolonií a především přežívání jejich U a L subpopulací. Zaměřila jsem se na kinázy Sch9p a Rim15p s centrálním postavením v signalizaci, transkripční aktivátory stresové odpovědi Msn2p a Msn4p a transkripční faktor Gis1p, který je důležitý pro přechod na respirační metabolismus. Mikroskopie tenkých řezů kolonií odhalila změnu ve vertikální diferenciaci kolonií kmene rim15Δ a částečně PTEF-SCH9 a změněnou morfologii U a L buněk kolonií ostatních analyzovaných kmenů. Změněná hladina Sch9p, Rim15p, Msn2/4p a Gis1p výrazně ovlivnila schopnost přežívání U i L buněk v čase. On solid surfaces, laboratory strain Saccharomyces cerevisiae BY4742 forms horizontally and vertically stratified colonies. Central part of the colony differentiates into U and L cells occupying upper and lower colony regions, respectively. There are many morphological and physiological differences between these two cell types. However, molecular mechanisms of the vertical stratification are still largely unknown. In this study, I analyzed effects of the deletion and overexpression of Sch9p a Rim15p kinases with crucial role in nutrient sensing pathways, of transcription activators Msn2p and Msn4p involved i stress response and of post-diauxic shift transcription factor Gis1p in yeast colony differentiation and U and L cell survival. Microscopic analyses showed changes in morphology of cells in colonies of these strains and decreased ability of vertical stratification in colonies of the strain rim15Δ and partially in colonies of the strain PTEF-SCH9. Moreover, changed level of these proteins significantly affected long-term survival of U and L cells. Obtained data suggest an involvement of nutrient sensing pathways in colony formation and development.
