Jejich použití je však omezeno kardiotoxicitou. Jediným doposud registrovaným léčivem schopným omezit kardiotoxicitu antracyklinů je dexrazoxan. Mechanismus jeho kardioprotektivního působení, stejně tak jako mechanismus antracyklinové kardiotoxicity zůstávají i přes půlstoletí výzkumu nejasné. V této práci jsme se soustředili na studium molekulárních mechanismů tohoto poškození, zvláště pak s ohledem na mechanismy kardioprotekce pomocí dexrazoxanu. Neprokázali jsme, že v patogenezi antracyklinové kardiotoxicity hraje významnou roli glutathion a s ním spojený enzymový systém, a to jak v experimentech in vitro tak i při analýze tkáně myokardu v rámci in vivo modelu chronické antracyklinové kardiotoxicity. Dále jsme zjistili, že dexrazoxan je schopen zabránit antracyklinové kardiotoxicitě v našem in vitro modelu izolovaných neonatálních kardiomyocytů, avšak nechrání tyto buňky před toxickým poškozením modelovým oxidačním insultem peroxidem...

Anthracyclines are amongst the most widely used antineoplastic agents. Nevertheless, their use is limited by the risk of cardiotoxicity. Dexrazoxane has been the only approved cardioprotectant against anthracycline cardiotoxicity so far. Despite half a century of research, the mechanisms of its cardioprotective ability as well as the mechanisms of anthracycline toxicity are elusive. In this study, we focused on the study of the molecular mechanisms of dexrazoxane cardioprotection. We did not prove the principal role of glutathione and related antioxidant enzymes in the pathogenesis of anthracycline cardiotoxicity both in the in vitro experiments and in vivo in the samples of left ventricles from the well-established model of chronic anthracycline cardiotoxicity in rabbits.
