Obezita je úzce spojena s rozvojem inzulínové rezistence, ektopickým ukládáním tuku v játrech a svalech, jaterní steatózou a diabetem 2. typu. Tuková tkáň obézních jedinců produkuje zvýšené množství adipokinů, které se podílejí na vzniku inzulínové rezistence, a zároveň do tukové tkáně infiltrují makrofágy způsobující mírný chronický zánět. Benefiční účinky na úroveň zánětu v tkáni a na metabolizmus lipidů a cukrů v organismu mají n-3 polynenasycené mastné kyseliny (n3-PUFA). Výzkum negativních důsledků obezity a možnost zlepšení těchto dopadů probíhá i na laboratorních zvířatech. Vlivem podávání diet s vysokým podílem tuku je u nich indukována obezita a dále jsou definovány dopady obezity na metabolizmus. Ve své diplomové práci jsem zkoumala vliv různých vysokotukových diet a diety obsahující n-3 polynenasycené mastné kyseliny na homogenitu tukových dep z hlediska velikosti buněk a množství zánětu v tukové tkáni. Dále jsem pozorovala rozvoj obezity v čase v závislosti na délce podávání vysokotukové diety. Metody: Intraperitoneální glukózový toleranční test (IPGTT), orální glukózový toleranční test (OGTT), stanovení množství lipidových metabolitů v plazmě, stanovení triacylglycerolů v játrech a tukové tkáni, stanovení množství DNA v jaterní a tukové tkáni, histologické stanovení velikosti...
Obesity is closely related to insulin resistance development, ectopic fat accumulation in the liver and skeletal muscles, liver steatosis and type 2 diabetes mellitus. Adipose tissue of obese subjects produces increased quantity of adipokines, which are involved in development of insulin resistance, and in the same time macrophages causing low-grade inflammation are infiltrating into adipose tissue. Long-chain polyunsaturated fatty acids of the n-3 series have beneficial effects on adipose tissue inflammation and lipid and carbohydrate metabolic pathways in organism. There is lot of research ongoing on laboratory animals on negative consequences of obesity and possible improvement. Obesity is induced by diet with high fat content and the consequences are evaluated. I have analysed the influence of different high fat diets and a diet with long-chain polyunsaturated fatty acids of the n-3 series on homogeneity of adipose depots in the cell size and the level of inflammation. I observed gradual development of obesity due to administration of high fat diet to laboratory animals.
