cell swelling, hypotonic stress, increased extracellular K+ concentration, regulatory volume increase, decrease, K+ channels, aquaporins(CZ) Jednou ze závažných komplikací ischemického a traumatického poškození mozku je vývoj mozkového edému, který vzniká v důsledku narušení iontové homeostázy, což vede k uvolnění velkého množství K+ a neurotransmiterů do extracelulárního prostoru. Zvýšený příjem osmoticky aktivních látek, který je zprostředkován především astrocyty, je kompenzován intracelulární akumulací vody, která rychle proniká do astrocytů prostřednictvím aquaporinu-4 (AQP4). Bylo prokázáno, že změna lokalizace AQP4 v membráně astrocytů prostřednictvím delece genu pro α-sytrofin, jehož protein je zodpovědný za lokalizaci AQP4 v perivaskulárních membránách astrocytů (Neely et al. 2001), má za následek snížení edemického nárůstu mozkové tkáně a pomalejší vychytávání K+ z extracelulárního prostoru (Amiry-Moghaddam et al. 2003). Cílem práce bylo objasnit, jak delece genu pro α-sytrofin ovlivní objemové změny kortikálních astrocytů v průběhu jejich vystavení modelovým patologickým stavům, jako je hypoosmotický a hyperosmotický stres, zvýšená koncentrace extracelulárního K+ (10mM K+ ) a deprivace glukózou a kyslíkem (OGD). Pro vizualizaci jednotlivých astrocytů, které postrádají protein α-syntrofin, jsme křížením GFAP/EGFP myší a α-syntrofin deficientních (α-Syn-/- ) myší vytvořili nový dvojitě- transgenní kmen GFAP/EGFP/α-Syn-/-...
(EN) The formation of brain oedema, which accompanies ischemic or traumatic brain injuries, originates from a disruption of ionic/neurotransmitter homeostasis that leads to extracellular K+ elevation and neurotransmitter accumulation in the extracellular space. An increased uptake of these osmotically active substances, predominantly provided by astrocytes, is accompanied by intracellular water accumulation via aquaporin-4 (AQP4). Since it has been shown that the removal of perivascular AQP4 via the deletion of α- syntrophin, which is the protein responsible for anchoring AQP4 on the astrocytic membrane (Neely et al. 2001), delays oedema formation and K+ clearance (Amiry-Moghaddam et al. 2003), we aimed to elucidate how the alpha-syntrophin deletion affects astrocyte volume changes in the cortex during pathological states, such as hypoosmotic stress or oxygen- glucose deprivation (OGD), using three-dimensional (3D) confocal morphometry in situ.
