Buněčný cyklus, zahrnující buněčné dělení a přípravu buněk na tento energeticky a regulačně extrémně náročný proces, má stěžejní význam pro zachování integrity těl všech mnohobuněčných organismů a pro udržení kontinuity života na Zemi. Porozumět tomu, jak je zajišťována regulace průběhu tohoto procesu, má zásadní význam pro pochopení fungování celé řady dalších procesů probíhajících v rostlinách i živočiších. Jedním z procesů souvisejících s buněčným cyklem, jehož význam pro lidstvo je z ekonomického hlediska nedozírný je tuberizace lilku bramboru (Solanum tuberosum L.). Brambor patří do čtveřice nejdůležitějších zemědělských plodin (spolu s kukuřicí, rýží a pšenicí - FAO 1995) a je pěstován a konzumován jako potravina ve většině zemí světa. Vhodným způsobem, jak lze proniknout do tajů regulace buněčného cyklu je transformace rostlin některým z genů, o nichž je známo, že v tomto procesu figurují. Této myšlenky využívá již několik let trvající studium rostlin tabáku (Nicotiana tabacum L.) transformovaných mitotickým aktivátorem cdc25 ze Schizosaccharomyces pombe (Spcdc25), kterému se tým Doc. H. Lipavské. (PřF UK, ČR) ve spolupráci s týmem Dr. D. Francise (Cardiff University, Great Britain) věnuje již řadu let. Tuberization is an important process in potato that is governed by a complex of environmental as well as internal factors. As a morphogenic process, it is closely connected with regulation of frequency and orientation of cell division. Thus, the aim of this thesis was to transform potato with mitotic activator cdc25 from Schizosaccharomyces pombe (Spcdc25) and to follow the resulting changes in potato phenotype with a special focus on tuber formation. In previous studies with tobacco, it has been shown that the expression of the above mentioned Spcdc25 gene results in dramatic changes in morphology (leaf structure, restricted root development and growth, and also remarkable developmental change - acceleration of flowering). Moreover, many other characteristics that can be induced in control by cytokinin application have been observed (e.g. cytokinin independent de novo shoot formation, carbohydrate metabolism changes). The two last characteristics - cytokinins independency of morphogenic process and changes in carbohydrate metabolism (e.g. increased starch deposition) - are important from the point of view of expected changes in characteristic of Spcdc25 transgenic potato.
