2 1 SOUHRN Hlavním faktorem řídícím glukózou stimulovanou inzulínovou sekreci (GSIS) v buňkách je pravděpodobně zvýšení ATP/ADP poměru následkem zvýšené glykolýzy a následně i oxidativní fosforylace. Bioenergetika INS-1E buněk, představujících platný model pro studium insulinové sekrece in vitro, byla kvantifikována pomocí respirometrie a současného snímání mitochondriálního membránového potenciálu ( m). Ukázali jsme, že přidání glukózy k buňkám, které ji vypotřebovaly, má za následek skokové zvýšení jak respirace, tak m během GSIS. Současně s tím se také zvýšil endogenní respirační poměr stavu 3/4, jehož závislost na glukóze byla hyperbolická. Maximální hodnoty dosahoval tento poměr při maximální GSIS. Zároveň jsme se pokusili postihnout "toxický" efekt mastných kyselin na inzulínovou sekreci tím, že byla studována GSIS za přídavku linoleové kyseliny. Tento přídavek způsobil podstatné zmenšení skoku respirace i m po přídavku glukózy, současně s tím snížil závislost poměru stavu 3/4 na glukóze a také množství sekretovaného inzulínu, což bylo způsobeno mitochondriálním odpřažením. Energetický stav mitochondrií, a tím i míra oxidativní fosforylace může být popsána bioenergetickými parametry. Kromě nich se tento stav také odráží ve struktuře mitochondriální sítě. - 2 - 1 SUMMARY Increased ATP/ADP ratio resulting from enhanced glycolysis and oxidative phosphorylation represents a plausible mechanism controlling the glucose-stimulated insulin secretion (GSIS) in pancreatic -cells. Using high resolution respirometry and parallel mitochondrial membrane potential ( m) monitoring we have quantified bioenergetics in rat insulinoma INS-1E cells representing a suitable model for studies of insulin secretion in vitro. Upon glucose addition to glucose-depleted cells, we have demonstrated a simultaneous increase in respiration and m during GSIS and shown that the endogenous state 3/state 4 respiratory ratio hyperbolically increases with glucose, approaching the maximum oxidative phosphorylation rate at maximum GSIS. Attempting to assess the basis of the "toxic" effect of fatty acids on insulin secretion, GSIS has been studied after linoleic acid addition. The linoleic acid addition diminished the observed respiration increase, m jump, and magnitude of insulin release, and reduced state 3/state 4 dependencies on glucose, which was caused by mitochondrial uncoupling.
