Disertační práce se zabývala aplikacemi zobrazovací a metabolomické hmotnostní spektrometrie v klinické chemii. Teoretická část disertační práce byla věnována samotné technice hmotnostní spektrometrie a jejímu využití v klinické chemii s důrazem na její tři typy použité při vypracování práce: hmotnostně-spektrometrické zobrazování, hmotnostní spektrometrie ve spojení s iontovou mobilitou a hmotností spektrometrie ve spojení s kapalinovou chromatografií. Následují tři kapitoly praktické části obsahující vlastní teoretické úvody (mimo oblast hmotnostní spektrometrie), experimentální části a výsledky s příslušnými diskuzemi. První praktická část disertační práce byla zaměřena na hmotnostně-spektrometrické zobrazování za použití laserové desorpce/ionizace za účasti matrice. Pomocí této techniky byly zmapovány časově a regionálně specifické změny v hladinách polyaminů, aminokyselin a neurotransmiterů po hypoxicko-ischemickém (HI) poškození mozku známého jako novorozenecká HI encefalopatie. Ta je u novorozenců významnou příčinou mortality a morbidity s incidencí přibližně dva až tři případy na 1000 živě narozených dětí ve vyspělých zemích. Poškození bylo simulováno na krysím Rice-Vannucciho modelu s vzorkováním mozkové tkáně a mozkomíšního moku 12, 24 a 36 hodin po HI poranění. Pro citlivější detekci metabolitů...
The dissertation thesis dealt with the applications of imaging and metabolomic mass spectrometry in clinical chemistry. The theoretical part of the dissertation thesis was devoted to the mass spectrometry technique itself and its use in clinical chemistry, with an emphasis on the three types of mass spectrometry used in the dissertation: mass spectrometry imaging, ion mobility mass spectrometry and liquid chromatography-mass spectrometry. The theoretical part is followed by three chapters of the practical part containing their own theoretical introductions (outside the area of mass spectrometry), experimental parts, results, and discussions. The first practical part of the dissertation was focused on mass spectrometry imaging using matrix-assisted laser desorption/ionization. Using this technique, time- and region-specific changes in polyamine, amino acid, and neurotransmitter levels were mapped upon hypoxic-ischemic (HI) brain injury known as neonatal HI encephalopathy. It is a significant cause of mortality and morbidity in newborns, with an incidence of approximately 2-3 cases per 1000 live births in developed countries.
