Xenopus laevis, regeneration, wound healing, gene expression, ECM, model organism, DPCA, qPCRRegenerace tkání představuje komplexní biologický proces, jehož studium může přispět k lepšímu porozumění mechanismů obnovy poškozených struktur a potenciálně i k rozvoji přístupů regenerativní medicíny. Obojživelník Xenopus laevis patří mezi významné modelové organismy pro studium regenerace, protože jeho embrya a larvy vykazují schopnost regenerovat ocas po amputaci, přičemž během vývoje dochází k přechodnému období snížené regenerační schopnosti, označovanému jako refrakterní fáze. Cílem této práce bylo analyzovat vliv vybraných látek na regeneraci ocasu u embryí X. laevis a charakterizovat vybrané molekulární mechanismy spojené s tímto procesem. V experimentální části byl sledován účinek látek DPCA (zmírňující zajizvení tkáně) a resolvinu D5 (snížení imunitní odpovědi) na regeneraci ocasu u embryí ve stadiích s rozdílnou regenerační schopností. Regenerace byla hodnocena po amputaci ocasu, kultivaci embryí v přítomnosti testovaných látek a následné morfologické analýzy. Dále byla analyzována genová exprese vybraných markerů spojených s buněčnou odpovědí na poranění a remodelací extracelulární matrix pomocí kvantitativní PCR. Tissue regeneration is a complex biological process, and its study can help to better understand the mechanisms involved in the repair of damaged structures, as well as contribute to the development of regenerative medicine approaches. The amphibian Xenopus laevis is an important model organism for studying regeneration, because its embryos and larvae are able to regenerate their tail after amputation. However, during development, there is a temporary period of reduced regenerative capacity, known as the refractory phase. The aim of this thesis was to analyze the effect of selected compounds on tail regeneration in X. laevis embryos and to describe selected molecular mechanisms associated with this process. In the experimental part, the effects of DPCA (a compound that reduces tissue scarring) and Resolvin D5 (which modulates the immune response) were tested on tail regeneration in embryos at developmental stages with different regenerative abilities. Regeneration was evaluated after tail amputation, followed by cultivation of embryos in the presence of the tested compounds and subsequent morphological analysis.
