mTORC2, oocyte, meiotic maturation, AKT, Rictor, cytoskeleton, in vitro fertilizationTato diplomová práce se zabývá rolí signální dráhy mTORC2 během meiotické maturace savčích oocytů. Cílem práce bylo objasnit vliv mTORC2 na průběh maturace oocytů, organizaci cytoskeletu a jejich vývojový potenciál. Analýza míry exprese proteinů ukázala rozdíly ve fosforylaci RICTOR a AKT mezi oocyty ve stádiu germinálního váčku (GV) a metafáze II (MII), což poukazuje na změny v aktivitě mTORC2 v průběhu meiotické maturace. Funkční význam této dráhy byl dále studován pomocí specifické farmakologické inhibice mTORC2 během in vitro maturace oocytů. Bylo prokázáno, že inhibice mTORC2 vede k signifikantnímu snížení podílu MII oocytů. Současně docházelo ke zpomalení průběhu meiotického dělení, snížení fosforylace AKT na serinu 473 a k poruchám organizace meiotického dělícího vřeténka a uspořádání chromozomů. MII oocyty vystavené mTORC2 inhibitoru vykazovaly rovněž snížený vývojový potenciál, zejména v raných stadiích embryonálního vývoje po in vitro fertilizaci. Získané výsledky ukazují, že mTORC2 hraje důležitou roli v regulaci meiotické maturace oocytů, pravděpodobně prostřednictvím ovlivnění buněčného cyklu a cytoskeletální organizace, a tím přispívá k zajištění jejich vývojové kompetence.
This thesis focuses on the role of the mTORC2 signalling pathway during meiotic maturation of mammalian oocytes. The aim of this study was to analyse the role of mTORC2 in oocyte maturation and developmental competence. Analysis of protein expression revealed stage-specific differences in the phosphorylation of RICTOR and AKT between oocytes at the germinal vesicle (GV) and metaphase II (MII) stages, indicating dynamic changes in mTORC2 activity during meiotic maturation. The functional significance of this pathway was further investigated using pharmacological inhibition of mTORC2 during in vitro maturation. Inhibition of mTORC2 resulted in a significant reduction in the proportion of MII oocytes, accompanied by delayed meiotic progression, reduced AKT phosphorylation at Ser473, and defects in spindle organization and chromosome alignment. Moreover, oocytes exposed to the mTORC2 inhibitor displayed impaired developmental competence, particularly during the early stages of embryonic development following in vitro fertilization. These findings suggest that mTORC2 plays a role in regulating oocyte meiotic maturation, likely through its involvement in cell cycle control and cytoskeletalorganization, therebycontributingto the establishmentofoocyte developmental competence.
