oocyte, nanoplastics, meiotic maturationMeiotické zrání představuje klíčovou fázi vývoje savčích oocytů, která může být nepříznivě ovlivněna environmentálními perzistentními látkami, včetně nanoplastů (NPs). NPs byly detekovány v řadě lidských tkání a biologických tekutin, včetně folikulární tekutiny žen. Vzhledem k jejich schopnosti bioakumulace v organismu, včetně reprodukčních orgánů savců, je nezbytné detailně zkoumat jejich vliv na reprodukční procesy. Současně je důležité zaměřit se na účinky nízkých koncentrací NPs, kterým jsou organismy reálně vystaveny. Tato práce patří mezi první, které popisují nepříznivé účinky nízkých koncentrací polystyrenových nanoplastů (PS-NPs) na specifické markery meiotického zrání prasečích oocytů in vitro. Na základě provedených experimentů nebyl prokázán negativní vliv PS-NPs na schopnost oocytů dosáhnout stadia metafáze II ani na hladinu mitochondriálního proteinu Mitofuzin 2. Naopak byl potvrzen jejich negativní vliv na zvýšený výskyt abnormalit meiotického vřeténka, změny hladin mitochondriálních proteinů MIRO1 a Sirtuin 3 a na epigenetické modifikace H3K4me2 a H3K9me3. Souhrnně tyto výsledky naznačují, že i nízké dávky PS-NPs mohou negativně ovlivňovat kvalitu oocytů, což může mít významné důsledky pro reprodukční schopnost samic.
Meiotic maturation represents a key stage of mammalian oocyte development, which can be adversely affected by persistent environmental substances, including nanoplastics (NPs). NPs have been detected in various human tissues and biological fluids, including follicular fluid in women. Given their capacity for bioaccumulation in the organism, including mammalian reproductive organs, it is essential to investigate their impact on reproductive processes in detail. Simultaneously, it is important to focus on the effects of low NPs concentrations to which organisms are realistically exposed, as most previous studies have utilized significantly higher concentrations. This work is among the first to describe the adverse effects of low concentrations of polystyrene nanoplastics (PS-NPs) on specific markers of porcine oocyte meiotic maturation in vitro. Based on the experiments performed, no negative effect of PS-NPs was demonstrated on the ability of oocytes to reach the metaphase II stage or on the level of the mitochondrial protein Mitofusin 2. Conversely, their negative impact was confirmed regarding the increased incidence of meiotic spindle abnormalities, changes in the levels of mitochondrial proteins MIRO1 and Sirtuin 3, and on epigenetic modifications H3K4me2 and H3K9me3.
