Staphylococcus aureus, biofilm, antibiotic rezistance, ica operon, agr operon, sarA1 Abstrakt Biofilm představuje klíčový faktor patogeneze a perzistence bakterií rodu Staphylococcus, včetně Staphylococcus aureus, jedné z nejvýznamnějších patogenních bakterií. Tato bakalářská práce má formou literární rešerše shrnout současné poznatky o genetických determinantách a regulačních mechanismech, které se podílejí na tvorbě biofilmu u S. aureus. Pozornost bude věnována zejména genům zapojeným do zprostředkování adheze, syntézy biofilmové matrix a systémům jejich regulace (např. operon ica, regulační dráhy agr a sarA). Práce dále diskutuje souvislosti mezi tvorbou biofilmu, antibiotickou rezistencí a virulencí stafylokoků a nastiňuje význam těchto poznatků pro klinickou praxi a možnosti terapie biofilmových infekcí. 1 Abstract Biofilm is a key factor in the pathogenesis and persistence of bacteria of the genus Staphylococcus, especially Staphylococcus aureus. This bachelor thesis aims to summarize current knowledge about the genetic determinants and regulatory mechanisms involved in biofilm formation in S. aureus in the form of a literature review. Particular attention will be paid to genes involved in adhesion, biofilm matrix synthesis, and regulatory systems (e.g., the ica operon, agr and sarA regulatory pathways). The thesis further discusses the links between biofilm formation, antibiotic resistance, and staphylococcal virulence, and outlines the significance of these findings for clinical practice and the possibilities for treating biofilm infections.
