Ultra-High Performance Liquid Chromatography (UHPLC), Candesartan cilexetile, Lercanidipine hydrochloride, Limit of Detection (LOD), Limit of Quantification (LOQ), Forced Degradation study, Drug Stability, Optimization of Chromatographic ConditionsTato diplomová práce je zaměřena na vývoj UHPLC metody pro současné stanovení Candesartanu cilexetilu a Lercanidipinu hydrochloridu včetně jejich degradačních nečistot ve dvouvrstvých tabletách. Separace probíhala na koloně typu C18 v gradientovém režimu s mobilní fází tvořenou fosfátovým pufrem a acetonitrilem a detekcí pomocí PDA. Během vývoje byly optimalizovány klíčové chromatografické parametry, zejména pH mobilní fáze, typ kolony, teplota, průtok a gradient eluce, s cílem dosažení dostatečné selektivity a časové efektivity metody. Vyvinutá metoda byla validována v souladu s požadavky ICH Q2(R1). Byla prokázána výborná linearita v rozsahu koncentrací 20-130 % (r ≥ 0,9998), vysoká přesnost (RSD ≤ 0,4 % pro Candesartan a ≤ 1,4 % pro Lercanidipin) a správnost v rozmezí 98-102 % pro stanovení obsahu. Metoda také prokázala dostatečnou robustnost a stabilitu analytických roztoků po dobu nejméně 48 hodin. Pro stanovení nečistot byla potvrzena dostatečná citlivost (LOQ ≤ 0,05 %). Součástí práce byla studie nucené degradace, která ukázala rozdílné degradační chování obou účinných látek. Candesartan byl citlivější vůči hydrolytickým podmínkám, zatímco Lercanidipin vykazoval vyšší náchylnost ke kyselé, oxidační a fotodegradaci. This diploma thesis is focused on the development of a UHPLC method for the simultaneous determination of Candesartan cilexetil and Lercanidipine hydrochloride including their degradation impurities in bilayer tablets. The separation was performed on a C18 column in gradient mode with a phase consisting of phosphate buffer and acetonitrile and detection using PDA. During the development, key chromatographic parameters, especially the pH of the mobile phase, column type, temperature, flow rate and elution gradient, were optimized in order to achieve sufficient selectivity and time-effectiveness of the method. The developed method was validated in accordance with the requirements of ICH Q2(R1). Excellent linearity in the range of 20-130% (r ≥ 0.9998), high precision (RSD ≤ 0.4% for Candesartan and ≤ 1.4% for Lercanidipine) and accuracy in the range of 98-102% for content determination were demonstrated. The method also demonstrated sufficient robustness and stable solutions for at least 48 hours. Sufficient sensitivity was confirmed for the determination of the impurity (LOQ ≤ 0.05%).
