LC-MS, MS, orexin A fragment, rat plasmaDiplomová práce se zabývá vývojem a validací selektivní LC-MS/MS metody pro stanovení specifického fragmentu orexinu A (17-33) v krysí plazmě. Orexiny jsou klíčové neuropeptidy regulující cyklus spánek-bdění a jejich deficit je hlavní příčinou narkolepsie 1. typu. Práce vznikla ve spolupráci s 1. LF UK za účelem objasnění farmakokinetiky tohoto fragmentu, jehož podávání představuje potenciální terapeutický přínos oproti intaktnímu neuropeptidu. Experimentální část byla zaměřena na optimalizaci separačních a detekčních parametrů na systému kapalinové chromatografie s tandemovým hmotnostním detektorem. K separaci analytu byla použita kolona InfinityLab Poroshell 120 EC-C18 (2,1 × 50 mm, 1,9 µm) od společnosti Agilent Technologies. Mobilní fáze se skládala z 0,1% kyseliny mravenčí (složka A) a methanolu (složka B). Separace probíhala za podmínek gradientové eluce (čas: 0,0-2,0-5,0-5,5-6,0-8,5 min, A: 80-80-0-0-80-80 % v/v) při průtoku 0,45 ml·min−1 . Teplota kolony byla nastavena na 40 řC, objem nástřiku činil 2 µl a teplota v automatickém dávkovači byla temperována na 15 řC. Identifikace analytu byla realizována v pozitivním ionizačním módu (ESI+) v režimu MRM. This master's thesis describes the development and validation of a selective LC-MS/MS method for the determination of a specific orexin A fragment (17-33) in rat plasma. Orexins are key neuropeptides that regulate the sleep-wake cycle; their deficiency is the primary cause of type 1 narcolepsy. This work was conducted in collaboration with the First Faculty of Medicine, Charles University, to clarify the pharmacokinetics of this fragment, as its administration offers potential therapeutic benefits compared to the intact neuropeptide. The experimental part focused on the optimization of separation and detection parameters using a liquid chromatography system coupled with a tandem mass spectrometer. An InfinityLab Poroshell 120 EC-C18 (2.1 × 50 mm, 1.9 µm) column from Agilent Technologies was used for analyte separation. The mobile phase consisted of 0.1% formic acid (eluent A) and methanol (eluent B). Separation was performed under gradient elution conditions (time: 0.0-2.0-5.0-5.5-6.0-8.5 min, A: 80-80-0-0-80-80 % v/v) at a flow rate of 0.45 ml·min−1 .
