Hlavním pacemakerem tohoto systému jsou suprachiasmatická jádra (SCN) hypotalamu, která přijímají fotickou informaci prostřednictvím retinohypotalamického traktu (RHT). Na této dráze působí glutamát jako klíčový excitační neurotransmiter a hlavní mediátor fotické synchronizace. Tato práce shrnuje současné poznatky o glutamátergické neurotransmisi mezi RHT a SCN a o mechanismech, které zprostředkovávají fázovou odpověď cirkadiánního oscilátoru na krátký světelný podnět. V úvodních kapitolách je popsána organizace cirkadiánního systému a experimentální důkazy glutamátergické povahy retinohypotalamického vstupu do SCN. Podrobněji jsou analyzovány mechanismy přenosu fotické informace z RHT na neurony SCN, včetně podjednotkového složení glutamátových receptorů a astrocytární regulace glutamátergické transmisie. Synaptická aktivace glutamátových receptorů v SCN vede k intracelulární signalizaci, která propojuje vstup z RHT s molekulárními hodinami v SCN. The circadian system of mammals synchronizes physiological and behavioural processes with the 24-hour light-dark cycle. The main pacemaker of this system are the suprachiasmatic nuclei (SCN) of the hypothalamus, which receive photic information via the retinohypothalamic tract (RHT). Along this pathway, glutamate acts as a key excitatory neurotransmitter and the principal mediator of photic entrainment. This thesis summarizes current knowledge on glutamatergic neurotransmission between the RHT and the SCN and on the mechanisms that mediate the phase response of the circadian oscillator to a brief light pulse. The introductory chapters describe the organization of the circadian system and experimental evidence for the glutamatergic nature of the retinohypothalamic input to the SCN. Subsequent sections provide a more detailed analysis of the mechanisms by which photic information carried by the RHT is transmitted to SCN neurons, including the subunit composition of glutamate receptors and astrocytic regulation of glutamatergic transmission. Synaptic activation of glutamate receptors in the SCN triggers intracellular signalling that links input from the RHT to the molecular clockwork in the SCN.
