Lipidomika je dynamicky se rozvíjející vědní obor, který se zaměřuje na studium struktury, funkce a vzájemných interakcí lipidových molekul v biologických systémech. Získané poznatky mohou významně přispět k vývoji nových terapeutik, biomarkerů a diagnostických metod, zejména v kontextu metabolických, kardiovaskulárních a neurodegenerativních onemocnění. Lipidy představují vysoce heterogenní skupinu látek, často se vyskytující ve formě izomerů a izobarů, což činí jejich strukturní charakterizaci značně náročnou. Kromě toho analýzu komplikují složitost biologických matric, nízké koncentrace některých lipidů, a v některých případech i omezené množství dostupného vzorku. Přestože v posledních letech došlo k významnému pokroku v oblasti hmotnostní spektrometrie a souvisejících analytických metod, vývoj nových technik a postupů nadále pokračuje. Tato disertační práce se zaměřuje na vývoj a optimalizaci pokročilých analytických přístupů pro analýzu lipidů a přispívá tak k rozvoji lipidomických metod. Chemická ionizace za atmosférického tlaku (APCI) a fotoionizace za atmosférického tlaku (APPI) patří mezi techniky, které jsou stále běžně využívané pro ionizaci neutrálních lipidů. Lipidomics is a dynamic and rapidly evolving field that focuses on the study of the structure, function, and interactions of lipid molecules in biological systems. The knowledge gained through lipidomic research can significantly contribute to the development of new therapeutics, biomarkers, and diagnostic tools for metabolic, cardiovascular, and neurodegenerative diseases. However, lipids represent a highly complex class of biomolecules characterized by numerous isobars and isomers, which makes their structural characterization challenging. In addition, the complexity of biological matrices, low analyte concentrations, and limited sample amounts further complicate the analysis. Over the past few decades, substantial progress has been made in developing advanced analytical techniques, and innovation in this area continues to advance rapidly. The aim of this thesis is to develop new ionization and fragmentation techniques that will contribute to the advancement of lipidomic methodologies. Atmospheric pressure chemical ionization (APCI) and atmospheric pressure photoionization (APPI) are still commonly used techniques for the ionization of neutral lipids.
