mitochondria, retina, oxidative stress, antioxidant protection, macular degeneration, diabetic retinopathy, glaucomSítnice je energeticky vysoce náročná tkáň, jejíž funkce závisí na efektivitě mitochondrií. Největší hustota a aktivita těchto organel byla pozorována ve vnitřních segmentech čípků a tyčinek, kde probíhá oxidační fosforylace - proces nezbytný pro tvorbu ATP, který je spojený i s produkcí reaktivních forem kyslíku. Nadměrná tvorba těchto molekul vede k oxidačnímu stresu a poškození mitochondriální DNA, stejně jako jiných buněčných struktur. K omezení těchto škodlivých účinků slouží buněčné antioxidační systémy, které zahrnují enzymy a molekuly, jako je vitamin C, karotenoidy a koenzym Q10. Nerovnováha mezi tvorbou a odstraněním reaktivních forem kyslíku přispívá k rozvoji degenerativních onemocnění sítnice. Práce se zaměřuje na tři taková onemocnění - věkem podmíněnou makulární degeneraci, diabetickou retinopatii a glaukom. U všech dochází k narušení energetického metabolismu, zvýšené tvorbě reaktivních forem kyslíku a ztrátě retinálních buněk. Mitochondrie zde představují nejen klíčový cíl patologických procesů, ale také potencionální terapeutický cíl. Výzkumy na zvířecích a buněčných modelech naznačují, že ochrana mitochondrií může zpomalit progresi těchto onemocnění. The retina is high energy depending tissue whose function depends on the efficiency of mitochondria. The highest density and activity of these organelles has been observed in the inner segments of cones and rods, where oxidative phosphorylation takes place - a process necessary for ATP production. It is also associated with the production of reactive oxygen species. Excessive production of these molecules leads to oxidative stress and damage of mitochondrial DNA, as well as other cellular structures. Cellular antioxidant systems, which include enzymes and molecules such as vitamin C, carotenoids, and coenzyme Q10, serve to limit these harmful effects. An imbalance between the production and removal of reactive oxygen species contributes to the development of degenerative retinal diseases. Present work is focused on such three selected diseases: age-related macular degeneration, diabetic retinopathy, and glaucoma. All of these diseases involve disruption of energy metabolism, increased production of reactive oxygen species, and retinal cell loss. Mitochondria are not only a key place of pathological processes, but also they are a potential therapeutic target.
