Supernova, DNA, deoxyribozyme, thermostability, single-step selectionTáto diplomová práca sa zaoberá zvyšovaním termostability svetlo produkujúceho deoxyribozymu Supernova, ktorý katalyzuje chemiluminiscenčnú reakciu závislú od zinku. Na štrukturálnej úrovni boli pripravené varianty s predĺženou triple-helix doménou, obsahujúcou rôzne kombinácie C·G·G a T·A·A tripletov. This thesis focuses on increasing the thermostability of the light-producing deoxyribozyme Supernova, a zinc-dependent catalyst of chemiluminescent reactions. At the structural level, sequence variants were designed by extending the triple-helix domain with different combinations of C·G·G and T·A·A triplets. In parallel, environmental conditions including pH, Znš⁺ concentration, and temperature were systematically investigated to evaluate their combined influence on catalytic performance. Results from ligation assays, NMR spectroscopy, and differential scanning calorimetry demonstrated that the SN2+6mix variant retains higher activity at elevated temperatures and exhibits greater structural stability compared with the parental Supernova sequences. These findings establish a framework for designing thermostable deoxyribozymes with potential applications in biosensing and diagnostics under thermally challenging conditions. Key words: deoxyribozyme, Supernova, thermostability, DNA triplex, chemiluminescence
