Muscle Hypertrophy, mTOR, Akt, Rag-GTPases, MechanosensorsRůst svalové hmoty je složitý proces regulovaný mnoha signálními drahami. Mezi klíčové molekuly, které stojí na rozcestí této signalizace, patří komplexy tvořené mTOR kinázou, mTORC1 a mTORC2. Tyto komplexy integrují signály z několika klíčových drah, které odpovídají na růstové faktory, dostupnost aminokyselin a mechanickou zátěž svalu. Na základě této signalizace regulují komplexy mTOR syntézu proteinů, lipidů, nukleotidů a autofágické procesy. Zvýšená míra proteosyntézy se pak přímo podílí na svalovém růstu syntézou nových myofibrilárních proteinů. Cílem této práce je analýza současného poznání o těchto klíčových dráhách, které s mTOR komplexy interagují a průzkum jejich ústřední role ve svalové hypertrofii. Muscle hypertrophy is a complex process regulated by several signaling pathways. Among the key molecules, which stand at the crossroads of hypertrophy signaling, are the complexes of the mTOR kinase, mTORC1 and mTORC2. These complexes integrate signals from key pathways, which respond to growth factors, amino acid availability and mechanical stress. In response to these pathways, the mTOR complexes modulate the synthesis of proteins, lipids, nucleotides and autophagic processes. Increased levels of proteosynthesis directly contributes to muscle growth through the synthesis of new myofibrilar proteins. The goal of the thesis is to analyze current knowledge of the key signaling pathways, which interact with the mTOR complexes, and to explore their overarching role in muscle hypertrophy.
