V této diplomové práci byly studovány hydrogely na bázi 2-hydroxymethakrylátu (HEMA). Bylo sledováno jejich možné využití pro studium kinetiky uvolňování hydrofobního protinádorového léčiva paklitaxelu (PTX), s cílem přiblížit se podmínkám simulujících in vivo prostředí. Paklitaxel je léčivo se silnými vedlejšími účinky, kterým lze částečně zamezit vhodnou lokální aplikací, proto v této práci byla studována polylatidová (PLA) nanovlákna s inkorporovaným léčivem. Dále byl sledován vliv přídavku polyethylenglykolu (PEG) o molekulové hmotnosti 20 kDa a CD do struktury PLA nanovláken na profily uvolňování PTX do různých prostředí simulující in vivo prostředí. Pro sledování kinetiky uvolňování PTX byla použita analytická HPLC metoda s UV detekcí. Přídavkem PEG do PLA nanovláken bylo docíleno výrazně vyššího množství uvolněného hydrofobního léčiva, které se zároveň uvolňovalo delší dobu, ve srovnání s vlákny obsahující pouze PTX. Tento trend byl zřejmý jak u experimentů sledující uvolňovaní PTX do PBS, tak i do hydrogelových matric. Vliv přídavku CD na uvolňování PTX byl zřejmý pouze u sledování kinetiky uvolňování PTX do hydrogelových matric, kde došlo k většímu uvolnění PTX z těchto vláken. In this diploma thesis, hydrogels based on 2-hydroxymethacrylate (HEMA) were studied. Their possible use to study the release kinetics of the hydrophobic anticancer drug paclitaxel (PTX) was investigated in order to approach the conditions simulating in vivo environments. Paclitaxel is a drug with strong side effects, which can be partially prevented by appropriate local application. In this work, polylatide (PLA) nanofibers with incorporated drug was studied. Furthermore, the effect of the addition of polyethylene glycol (PEG) with a molecular weight of 20 kDa and CD to the structure of PLA nanofibers on the profiles of PTX release into various environments simulating in vivo environments was studied. An analytical HPLC method with UV detection was used to monitor the release kinetics of PTX. The addition of PEG to PLA nanofibers resulted in a significantly higher amount of hydrophobic drug released, which was also released over a longer period of time, compared to fibers containing only PTX. This trend was evident in both experiments monitoring the release of PTX into PBS and into hydrogel matrices.
