Trypanosoma brucei, iron, iron metabolism, iron-containing proteins, iron-sulfur clustersTrypanosoma brucei je parazitický organismus, který se nejčastěji vyskytuje v oblasti subsaharské Afriky a způsobuje spavou nemoc u člověka a onemocnění u zvířat. Na svého definitního savčího hostitele (člověka) je přenášen krevsajícími mouchami tse-tse (Glossina). Během svého složitého životního cyklu, kdy se vyskytuje v odlišných prostředích, prochází morfologickými a metabolickými změnami. Železo je důležitým prvkem pro všechny živé organismy, a tedy i pro Trypanosomy. Vzhledem k tomu, že jsou Trypanosomy závislé na příjmu železa z hostitele či přenašeče, hraje tento kov roli ve vztahu mezi parazitem a jeho hostitelem. Trypanosomy využívají železo v metabolických dějích, ať už se jedná o energetický metabolismus, respiraci, syntézu nukleových kyselin, detoxifikaci a buněčnou homeostázu. Je důležitým prvkem esenciálních drah tvorby takzvaných železo-sirných klastrů, které fungují jako kofaktory řady enzymů figurujících ve výše zmíněných procesech. Pochopení metabolismu železa v buňce může pomoci při vývoji nových léčiv a příkladem mohou být takzvané železné chelátory.
Trypanosoma brucei is a parasite most frequently occurring in Sub-Saharan Africa that causes sleeping sickness in humans and various similar illnesses in animals. The bloodsucking tsetse flies (Glossina) transfer the parasite to humans, their final hosts. Throughout its complex life cycle, Trypanosoma occurs in different environments and undergoes various morphological and metabolic changes. Iron is an important element for all living organisms, including Trypanosoma. The metal plays a crucial role in the host-parasite interaction since trypanosomes are dependent on the iron they acquire from the host or vector. Trypanosomes use iron in metabolic reactions, such as energy metabolism, respiration, nucleic acid synthesis, detoxification, and cellular homeostasis. It is an important element in the synthesis of iron-sulfur clusters which function as cofactors during the above-mentioned reactions. The understanding of iron metabolism in the cell can facilitate the development of new medicaments, an example being iron chelators.
