dc.description.abstract | Výzkum cytotoxicity titanocen dihalogenidů je tématem, které se objevuje v literatuře již od 70. let 20. století. Původně intenzivně studovaný titanocen dichlorid, který zklamal v 2. fázi klinických studií na lidech, byl dnes již nahrazen novější generací komplexů, které zpravidla nesou aryl-substituované cyklopentadienylové ligandy, a jejichž aktivitu lze dále podpořit substitucí chloridových ligandů fluoridy. Avšak mechanismus účinku těchto komplexů zůstává do značné míry záhadou, počínaje transportem do buněk, stabilitou ve fyziologickém prostředí, atd. V rámci této práce je připravena série různě substituovaných titanocen dichloridů a difluoridů, které jsou součástí větší série látek, která poslouží k bližšímu porozumění strukturních vlivů na cytotoxicitu těchto komplexů. Připravené látky jsou charakterizovány 1 H ,13 C NMR a IČ spektroskopií. Látky obsahující fluoridové ligandy pak i 19 F NMR spektroskopií, dále jejich bodem tání, a v případě dostupnosti monokrystalů také RTG strukturní analýzou a elementární analýzou. dc.description.abstract | Cytotoxicity of titanocene dihalides has been a persistent topic in scientific literature, dating back to the 1970's. The intensively studied titanocene dichloride, which has disappointed in phase II clinical trials on humans, has been substituted by various titanocenes, which bear aryl-substituted cyclopentadienyl ligands. The activity of these complexes can be interestingly further improved via substitution of chloride ligands by fluorides. However, the mechanism of action of these complexes remains somewhat of a mystery, starting with their transport into the cells, stability under physiological conditions, etc. This thesis describes the preparation of several variously substituted titanocene dichlorides and difluorides. These complexes are a part of a larger series, which will help to further the understanding of structural effects on the cytotoxicity of titanocenes. All prepared complexes are characterized via 1 H ,13 C NMR and IR spectroscopy. Complexes containing fluoride ligands have also been characterized via 19 F NMR spectroscopy, melting points, RTG structural analysis (if monocrystals were available) and elemental analysis.
