biodrying, sewage sludge, PLFA, microbial communitiesCílem této diplomové práce je využití analýzy fosfolipidových mastných kyselin (PLFA) pro stanovení skupin mikroorganismů v průběhu biosušení čistírenských kalů a jejich popis vzhledem k teplotnímu režimu biosušení. Byly sledovány 4 experimentální cykly A, B, C, D, přičemž cykly A, B a C, D se vzájemně lišily čistírenským kalem odebíraným ze dvou čistíren odpadních vod. Každý cyklus A, B, C, D probíhal ve 2 reaktorech s různými režimy provzdušňování pro porovnání mezofilního (max. teploty ±50 řC) a termofilního (max. teploty ±70 řC) režimu biosušení. Pro stanovení mikrobiálních skupin byla použita analýza fosfolipidových mastných kyselin (PLFA). Koncentrace jednotlivých PLFA byly podrobeny analýze hlavních komponent (PCA). Celková koncentrace PLFA odpovídající celkové mikrobiální biomase klesala v průběhu obou teplotních režimů cyklů B, C a D, oproti tomu ke konci obou teplotních režimů cyklu A byl zaznamenán nárůst celkové koncentrace PLFA. Výsledné celkové koncentrace PLFA dosahovaly statisticky vyšších hodnot na konci mezofilních režimů cyklů B, C a D. Vývoj poměru hub a bakterií ukázal, že houby se více uplatňovaly s postupem obou teplotních režimů cyklů A, B a termofilního režimu cyklu C. The aim of this thesis is the utilization of phospholipid fatty acids (PLFA) analysis to determine groups of microorganisms present during the biodrying of sewage sludge and their description in relation to the temperature regime of the biodrying. Four experimental cycles A, B, C, D were followed. Cycles A, B and C, D differred from each other in the sewage sludge collected from two waste water treatment plants. The cycles A, B, C and D were each performed in two reactors with different aeration regimes to compare the mesophilic (max. temperature ±50 řC) and thermophilic (max. temperature ±70 řC) regimes of biodrying. PLFA analysis was used to determine the microbial groups. Concentrations of individual PLFA were subjected to principal component analysis (PCA). The total PLFA concentration corresponding to total microbial biomass was decreasing during both temperature regimes of cycles B, C, D, while the total PLFA concentration increased towards the end of both temperature regimes of cycle A. The total PLFA concentration reached statistically higher values at the end of the mesophilic regime of cycles B, C and D.
