Galectins, β-N-acetylhexosaminidases, multivalent ligands, HPMA copolymers, recombinant expression, enzymatic synthesis, inhibitory potencialGalektiny jsou proteiny patřící do skupiny lektinů, které jsou schopné vázat sacharidové jednotky a dokážou specificky rozeznat glykany vystavené na povrchu buněk. Galektiny se in vivo účastní např. kancerogeneze, angiogeneze či fibrózy. Jejich výskyt signifikantně stoupá v souvislosti s řadou patogenních procesů, a proto je lze využít jako markery u některých typů rakovin nebo kardiopatologií a též pro cílenou vazbu terapeutik a/ nebo zobrazovacích činidel v diagnostice a terapii. Galektin-3 má specifickou strukturu označovanou jako chimerická a je schopný tvořit multivalentní oligomery. Přirozenými ligandy galektinů jsou glykany obsahující terminální β-galaktosidy, zvláště N-acetyllaktosamin, avšak vazba monovalentních glykanů je velmi slabá. Glykokonjugáty s vysokou afinitou ke galektinovým receptorům jsou optimálně multivalentní, biokompatibilní a stabilní in vivo. Tato kritéria splňují sacharidové ligandy konjugované na rozpustné a strukturně flexibilní N-(2-hydroxypropyl)methakrylamidové (HPMA) kopolymery. V rámci této práce byly pomocí enzymové syntézy připraveny dva druhy funkcionalizovaných disacharidů, založené na N-acetyllaktosaminu (Galβ4GlcNAc) a jeho strukturním analogu N,N'-diacetyllaktosaminu (GalNAcβ4GlcNAc). Galectins are proteins, wich belong to a group of lectins that are able to bind to saccharide units and they specifically recognize glycans exposed to the surface of the cells. Galectins participate in vivo, for example, in carcinogenesis, angiogenesis or fibrosis. Their occurrence increases significantly in connection with a number of pathogenic processes, therefore they can be used as markers for some types of cancer or cardiopathology and also for the targeted binding of therapeutics and/ or imaging agents in diagnosis and therapy. Galectin-3 has a specific structure known as chimeric and it is capable of forming multivalent oligomers. The natural ligands of galectins are glycans containing terminal β-galactosides, especially N-acetyllactosamine, but the binding of monovalent glycans is very weak. Glycoconjugates with high affinity to galectin receptors are optimally multivalent, biocompatible and stable in vivo. These criteria accomplish carbohydrate ligands conjugated to soluble and structurally flexible N-(2-hydroxypropyl)methacrylamide (HPMA) copolymers.
