Abstrakt: Použití bez značkových afinitních biosensorů, pokročilých implantátů a zařízení přicházejících do kontaktu s krví vyžaduje, aby jejich povrch odolával nespecifické adsorpci proteinů (fouling) a souvisejícím komplikacím a přitom aby měl na svém povrchu navázány bioreceptory poskytující specifickou funkci. Cílem této práce bylo prozkoumat strategie pro přípravu těchto bioaktivních povrchů potlačujících nespecifické interakce. Byly připraveny různé typy polymerních kartáčů, u kterých byla vyhodnocena jejich schopnost potlačovat nespecifické interakce po kontaktu s krevní plasmou (získanou od různých dárců a komerčních zdrojů) a vznik trombu po kontaktu s plnou krví. Pomocí silové spektroskopie byla dále stanovena energie a síla adheze bakterií, která byla u polymerních kartáčů významně snížena ve srovnání s různými referenčními substráty. Imobilizace bioreceptorů byla řešena strategií hierarchické funkcionalizace. Polymerní kartáče se skládaly z inertního spodního bloku a reaktivního horního bloku laditelné tloušťky. The aim of this thesis was to explore the strategies for the preparation of these antifouling bioactive surfaces. Various types of recently developed antifouling polymer brushes were synthesized and evaluated in terms of their ability to prevent fouling from blood plasma (obtained from various commercial sources and individual donors) as well as thrombus formation when contacted with whole blood. Moreover, single-cell force spectroscopy was utilized to characterize the bacterial adhesion force and energy on these polymer brushes, which were significantly reduced in comparison to various reference substrates.
