Barbora Szotáková, Ph.D. Školitel specialista: RNDr. Věra Králová, Ph.D. Název rigorózní práce: Vliv flubendazolu na proliferaci buněk kolorektálního karcinomu in vitro Flubendazol je široce užívané anthelmintikum patřící do skupiny benzimidazolů. Molekulární mechanismus účinku flubendazolu spočívá ve specifické vazbě na tubulin, která naruší strukturu a funkci mikrotubulů a mikrotubuly zprostředkovaný transport sekrečních váčků v absorpčních tkáních helmintů. Tyto vlastnosti benzimidazolových derivátů, narušovat mikrotubuly, nedávno zvýšily zájem o použití těchto látek jako možných protinádorových prostředků. Z dostupných studií a farmakologických údajů se dá předpokládat, že po perorálním podání flubendazol dosahuje vysoké koncentrace v buňkách tlustého střeva, proto jsme provedli studie ke zjištění potenciálního antiproliferativního účinku na liniích nádorových střevních buněk. Buněčné linie kolorektálního karcinomu SW480, SW620 a linie NCM460 pocházející z buněk normální střevní sliznice jsme ovlivnili rozdílnou koncentrací flubendazolu (0,1 - 10 µM) a následně inkubovali po dobu 24, 48 a 72 hod. The molecular mechanism of action of flubendazole is based on specific binding to tubulin, which results in disruption of microtubule structure and function and in the interference with the microtubule-mediated transport of secretory vesicles in absorptive tissues of helminths. The microtubule-disrupting properties of benzimidazole derivatives raised recently interest in these compounds as possible anti-cancer agents. Because of available studies and pharmacological statements, it is supposed considerable concentrations of flubendazole in intestinal cells after p.o. administration could be reached, we performed this study to investigate the antiproliferative potential of flubendazole in a panel of intestinal cancer cell lines.
