Barbora Szotáková, Ph.D. Název disertační práce: IDENTIFIKACE NOVÝCH FAKTORŮ VIRULENCE INTRACELULÁRNÍHO PATOGENA FRANCISELLA TULARENSIS Předložená disertační práce je zaměřena na studium úlohy bakteriálních tetratricopeptide repeat-like (TPR-like) proteinů v patogenesi infekce. Modelovým organismem byla zvolena vysoce virulentní intracelulární bakterie Francisella tularensis, jejíž mechanismus patogenese není doposud plně objasněn. V první části disertační práce byly na základě bioinformatických studií identifikovány tři geny (FTS_0201, FTS_1680, a FTS_0778) s predikovanou TPR-like doménou. Pro přípravu mutantních kmenů vybraných genů byla využita mutagenese TargeTron. Připravené mutantní kmeny byly použity pro studium role vybraných proteinů v patogenitě a imunogenitě F. tularensis s využitím in vitro a in vivo modelů a dále pro studium zapojení těchto proteinů v adaptačních mechanismech na stres. Z výsledků disertační práce bylo zjištěno, že protein FTS_1680 je významný pro intracelulární replikaci a plnou virulenci bakterie. Barbora Szotáková, Ph.D. Title of Doctoral Thesis: IDENTIFICATION OF NEW VIRULENCE FACTORS IN INTRACELULLAR PATHOGEN FRANCISELLA TULARENSIS This dissertation thesis is focused on the role of bacterial tetratricopeptide repeat-like (TPR-like) proteins in the pathogenesis of infection. As a model organism we have chosen highly virulent intracellular bacterium Francisella tularensis (F. tularensis), whose mechanism of pathogenesis is not completely described. In the first part of dissertation thesis we took advantage of bioinformatic methods and identified three genes (FTS_0201, FTS_1680, and FTS_0778) with predicted TPR-like domains. Mutants defective in protein expression were prepared by TargeTron insertion mutagenesis. Prepared mutant strains were used for studying the role of selected proteins in pathogenicity and immunogenicity of F. tularensis subsp. holarctica strain employing in vivo and in vitro models and further for studying the involvement of these proteins in stress tolerance.
