Univerzita Karlova v Praze, Farmaceutická fakulta v Hradci Králové Katedra Farmaceutické technologie Kandidát Mgr. Pavla Šináglová Konzultant PharmDr. Jitka Mužíková, Ph.D. Název rigorózní práce Studium nového směsného suchého pojiva na bázi sprejově sušené laktosy a mikrokrystalické celulosy Cílem práce bylo porovnání lisovatelnosti a doby rozpadu tablet ze směsného suchého pojiva Disitequik™ MCC a fyzikálních směsí suchých pojiv, které obsahovaly sprejově sušenou laktosu FlowLac® 100 a mikrokrystalickou celulosu Microcel® MC-102 v poměrech 1:9, 2:8 a 3:7. K tabletovinám byla přidána mazadla stearan hořečnatý a poloxamer 407 v koncentracích 1% a 2%. Lisovatelnost byla hodnocena pomocí energetické bilance lisování a pevnosti tablet. Příprava tablet probíhala na materiálovém testovacím zařízení T1-FRO50 TS.S1K Zwick/roell. Disitequik™ MCC poskytuje pevnější tablety s delší dobou rozpadu než odpovídající fyzikální směs sprejově sušené laktosy s mikrokrystalickou celulosou, vykazuje i vyšší hodnoty celkové energie lisování, v důsledku vyšších hodnot energie akumulované tabletou a vyšší plasticitu.
Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical technology Candidate Mgr. Pavla Šináglová Consultant PharmDr. Jitka Mužíková, Ph.D. Title of Thesis Study of a new mixed dry binder based spray-dried lactose and microcrystalline cellulose The aim of this study was to compare compressibility and tablet disintegration time of tablets from co-procesed dry binder Disitequik™ MCC and physical mixture of dry binders containing spray-dried lactose FlowLac® 100 and microcrystalline cellulose Microcel® MC-102 in ratios of 1:9, 2:8 and 3:7. Lubricants magnesium stearate and poloxamer 407 at concentrations of 1% and 2% were added to the mixtures. Compressibility was evaluated using the energy profile of the compression and tenile strength of tablets. The tablets were prepared on the material testing device T1-FRO50 TS.S1K Zwick / Roell. Disitequik™ MCC had a stronger tablet with a longer disintegration time than the tablets from the physical mixtures of spray-dried lactose with microcrystalline celulose. The tablets from Disitequik™ MCC show higher values of total energy pressing, due to higher levels of energy stored in the tablets and higher plasticity.
