4 Abstrakt Celiakie je chronické střevní onemocnění autoimunitního charakteru, které se vyskytuje převážně u geneticky predisponovaných osob. U pacientů dochází k nepřiměřené imunitní odpovědi na přítomnost glutenu obsaženého v některých obilovinách. Pacienti jsou doživotně závlislí na dodržování přísného dietního režimu. Genetická predispozice je z velké části založena na přítomnosti alel HLA-DQ2 a HLA- DQ8. Zbytek genetického rizika dosud není objasněn, ale vzhledem k autoimunitnímu charakteru onemocnění lze předpokládat, že genetická predispozice by mohla být skryta v polymorfismech genů, které se podílejí na směrování či regulaci imunitní odpovědi. CÍLE: Našim cílem bylo zjistit korelaci mezi fenotypem a genotypem u členů rozsáhlé třígenerační rodiny s mnohočetným výskytem celiakie a dalších autoimunitních chorob a ověřit, které vybrané genotypy korelují s fenotypem a do jaké míry. U všech členů rodiny jsme provedli genotypizaci alel HLA-DQB1 a HLA-DRB1, dále genotypizaci polymorfismů C1858T genu pro PTPN22, C1188A genu pro IL12B a C2029T genu pro TLR2. U TLR2 byla sledována také míra exprese na úrovni proteinu a na úrovni mRNA. METODY: Genotypizace HLA-molekul byla provedena pomocí metody PCR se sekvenčně specifickými primery, genotypizace polymorfismů v genech pro PTPN22 a IL12B pomocí metody PCR-RFLP...
5 Abstract Coeliac disease is a chronic intestinal autoimmune desease that is diagnosed in genetically predisposed people. This disease is characterized by inadequate immune response to gluten, which is contained in some cereals. The patients have to keep a strict dietary regime. The genetic predisposition is based on possession allels HLA-DQ2 and HLA-DQ8. The remainder genetic risc is unknown, but with regard to autoimmune character of disease, it is supposed that genetic predisposition can be included in polymorphisms of genes, which participate in a route or regulation of immune response. AIMS: We studied a correlation between phenotype and genotype in members of three-generation family, which is affected of coeliac disease and other autoimmune diseases. We tried to verify a correlation between phenotype and some genotypes. We tested the HLA-DQB1 and HLA-DRB1 alleles, the polymorphisms C1858T in PTPN22 gene, A1188C in IL12B gene and C2029T in TLR2 gene. We measured an expression of TLR2 protein and TLR2 mRNA as well. METHODS: Testing of HLA-molecules was performed using the PCR-SSP technique.
