Práce je zaměřena na vývoj a aplikaci nových metod detekce DNA a její syntézy in situ. V jejím průběhu byly vyvinuty 3 metodické pří- stupy, které výrazně usnadňují detekce buněčné DNA a současně popsány mechanismy cytotoxického působení 2'-deoxy-5-etynyluridinu. Vyvinuté přístupy představují důležitý nástroj pro studium replikace a opravných mechanismů DNA, studium mitochondriální DNA a rov- něž analýzu buněčné proliferace. První část práce je věnována po- pisu nové metody detekce 5-bromo-2'-deoxyuridinu inkorporovaného do nově syntetizované DNA pomocí monovalentních iontů mědi v přítomnosti kyslíku. Tato metoda je využitelná jak v případě detekce jaderné syntézy, tak mitochondriální syntézy DNA. V další části je popsána metoda vysoce efektivní detekce jaderné a mitochondri- ální DNA. Třetí metoda se zabývá eliminací nespecifických vazeb pro- tilátek namířených proti 5-bromo-2'-deoxyuridinu s 2'-deoxy-5-etynyluridinem. Tuto metodu lze s výhodou využít ve studiích, ve kterých je nezbytné použití dvou po sobě jdoucích pulzních značení. Postup je založen na využití nefluorescenčních azido molekul. Závěrečná část práce je za- měřena na cytotoxicitu 2'-deoxy-5-etynyluridinu a jeho metabolický účinek. 1
The thesis is focused on the development and application of the new methods for targeting DNA and DNA synthesis in situ. Three methodical approaches were developed, which significantly improve detection of cellular DNA. Concurrently, a new insights in the mech- anism of 2'-deoxy-5-ethynyluridine cytotoxicity were described. The approaches developed in this work provide an important tool for study of DNA replication and repair mechanisms, the study of mito- chondrial DNA, as well as the analysis of cell proliferation. The first part of the thesis presents the new method for the detection of 5-bromo-2'-deoxyuridine incorporated into the newly synthesized DNA using monovalent copper ions in the presence of oxygen. This method is applicable for the detection of nuclear DNA synthesis as well as for the mitochondrial DNA synthesis. Next part of the work describes the approach enabling highly efficient detection of over- all nuclear and mitochondrial DNA. The third approach concerned the elimination of the unspecific binding of anti 5-bromo-2'-deoxyuridine antibodies to another modified nucleoside - 2'-deoxy-5-ethynyluridine.
