phenylboronic acids, fluorescence, carbohydratesPráce se zabývá vlivem fruktosy, glukoronové kyseliny, glukosy a manosy na fluorescenční charakteristiky sondy Click fluor pomocí stacionární a časově rozlišené fluorescenční spektroskopie. Měřila se detailně spektra pro široký rozsah koncentrací sacharidů (do 3M). Změna intenzity fluorescence po přidání sacharidů se využila pro výpočet vazné konstanty komplexu Click-fluor/sacharid. Pro glukosu a fruktosu se také změřily křivky dohasínání fluorescence. Posledním tématem byla vazba značky Click-fluor na poly(ethylen glykolový), PEG, polymerní řetězec s navázanými molekulami glukosy jako koncovými skupinami, glc- PEG-glc. Z měření vyplývá, že největší afinitu má značka Click-fluor na fruktosu, kdežto tvorba esteru s glukosou je velmi slabá. V emisním spektru byly pozorovány celkem tři pásy. Jeden odpovídá nenavázané značce, druhý nestabilní formě esteru s cukrem a třetí stabilní cyklické formě esteru s cukrem. V časově rozlišené fluorescenci značky Click-fluor sice byly patrné změny u vlnových délek odpovídajícím esteru s glukosou, ne však příliš výrazné. Z emisních spekter komplexu Click-fluor a polymer glc-PEG-glc. i z hodnot středních dob života fluorescence vyplývá, že dochází k relativně silné vazbě značky na glukosou modifikovaný polymer. This paper deals with influence of fructose, glucuronic acid, glucose and mannose on the fluorescence characteristics of probe Click fluor using stationary and time resolved fluorescence spectroscopy. The spectra were measured for a wide range of concentrations (up to 3M), and the change in fluorescence intensity after the addition of carbohydrate was used to calculate the binding constants of the Click fluor/saccharide complex. Also time resolved fluorescence was measured for glucose and fructose, The last topic was the linking of Click- fluor to poly(ethylene glycol), PEG, with glucose at the end of the chain, glc-PEG-glc. The measurements show that the affinity of the Click-fluor for fructose is the greatest, on the other hand formation of the ester with glucose is very weak. Three strips were observed in the emission spectrum. One corresponds to the unbound probe, the second unstable ester form with sugar, and the third stable cyclic form of the sugar ester. In the time-resolved fluorescence of Click-fluor, changes in the wavelengths corresponding to the carbohydrate ester were seen, but not too pronounced.
