binding protein, combinatorial library, selection technique, phage display, ribosome display, yeast displayUmelo vytvárané väzobné proteíny odvodené od malých proteínových domén lákajú čoraz väčšiu pozornosť ako alternatívy k monoklonálnym protilátkam a majú široké spektrum využitia. Svoju aplikáciu si našli v oblastiach diagnostiky rôznych onemocnení, sú prísľubom pre vývoj vakcín proti globálnym ochoreniam ako je AIDS, môžu mať terapeutický potenciál a skvalitňujú purifikačné a chromatografické metódy. Na selekciu proteínových variánt, ktoré majú najlepšie väzobné vlastnosti sa v súčasnosti využíva viac než desať selekčných, tzv. displejových techník. Displejové techniky využívajú rôzne vektory pre vystavovanie proteínových variánt ako je tomu napríklad u fágového, kvasinkového, retrovirálneho či bakulovirálneho displeja. Selekcia taktiež môže prebiehať in vitro. Príkladom je ribozomálny displej, mRNA displej a CIS displej. Evolúcia displejových selekčných techník umožnila selekciu postranslačne glykosylovaných, fosforylovaných či acylovaných proteínov, prispela k zvyšovaniu výťažku selektovaných ligandov a k skvalitneniu ich väzobných vlastností. Práca popisuje a porovnáva vybrané selekčné metódy, pojednáva o výhodách a limitáciach ich kombinatoriálnych knižníc peptidov a uvádza praktické využitie väzobných proteínov odvodených od malých proteínových domén.
Artificial binding proteins derived from small protein domains attract attention as a promising alternative to monoclonal antibodies and can be used in many kinds of applications. They are useful in diagnosis of human diseases, seem to be a clue for more efficient vaccine development preventing from global diseases such as AIDS, can exhibit a therapeutic potential or improve purification techniques. For the selection of protein variants with desired properties such as high specificity and binding affinity, more than 10 different selection techniques have been developed. So called display techniques such as phage display, yeast display, retroviral display or baculovirus display are based on protein expression from different vectors. Contrary that, ribosome display, mRNA display and CIS display are cell-free systems based on in vitro translation. Development of different selection approaches allows production of post- translationally glycosylated, phosphorylated and acetylated proteins, increased yield of the produced binders and improved their binding properties.
