Perivascular adipose tissue, vascular smooth muscle cells, atherosclerosis, cardiovascular diseaseAbnormální proliferace buněk hladké svaloviny cév (VSMC) hraje důležitou roli v patogenezi aterosklerózy. Adipocyty produkují řadu parakrinně působících bioaktivních látek, které mohou působit na růst a migraci VSMC. Naše studie je zaměřena na schopnost epikardiálních adipocytů produkovat bioaktivní látky s přímým efektem na míru proliferace VSMC. Genová exprese lidských cytokinů (IL-6, IL-8, IL-18, RANTES a MCP-1) a adipokinů (leptin a adiponektin) byly měřeny v primární buněčné kultuře epikardiálních a viscerálních adipocytů, obojí v případě nediferencovaného a maturovaného stavu buněk. Produkce adipokinů (IL-6, IL-8, MCP-1, VEGF a adiponektin) v kondiciovaných médiích byla měřena pomocí Luminex testu. Míra proliferace VSMC byla měřena po kokultivaci s médiem získaným z primárních kultur adipocytů. Epikardiální preadipocyty prokázaly zvýšenou expresi IL-8 (3,25krát, p<0,05) ve srovnání s viscerálními preadipocyty. Exprese adiponektinu v epikardiálních preadipocytech byla výrazně snížena ve srovnání s expresí ve viscerálních preadipocytech (p<0,0001). Kromě toho genová exprese závisela na míře diferenciace a buněčném původu. Pozorovali jsme změněnou expresi IL-8 (0,016krát, p<0,01) a MCP-1 (0,19krát, p<0,05) v diferencovaných epikardiálních adipocytech oproti nediferencovaným adipocytům. Prokázali...
Abnormal vascular smooth muscle cell (VSMC) proliferation is thought to play an important role in the pathogenesis of atherosclerosis. Adipocytes produce several paracrine bioactive substances that can affect VSMC growth and migration. Our study focused on the ability of epicardial adipocytes to produce bioactive substances together with studying of direct effect of these substances on the VSMC proliferation rate. The gene expression of human cytokines (IL-6, IL-8, IL-18, RANTES and MCP-1) and adipokines (leptin and adiponectin) was measured in primary cell lines of epicardial and visceral adipocytes, both in undifferentiated and mature statuses. Moreover, adipokine production (IL-6, IL-8, MCP-1, VEGF and adiponectin) in conditioned media obtained from above mentioned primary cell cultures of adipocytes was measured by a Luminex assay. The VSMC proliferation rate was measured after co-culturing with CM obtained from primary cell cultures of adipocytes. The epicardial preadipocytes showed an increased expression of IL-8 (3,25-fold, p<0,05) compared with visceral preadipocytes.
