murine polyomavirus, VP2, VP3, minor capsid proteins, protein modification, protein-membrane interactionMyší polyomavirus (MPyV) je jedním z modelových zástupců čeledi Polyomaviridae. Kapsida MPyV se skládá z hlavního kapsidového proteinu VP1 a minoritních proteinů VP2 a VP3. Minoritní kapsidové proteiny pravděpodobně napomáhají přesunu virového genomu přes membránu endoplasmatického retikula na cestě do jádra v časné fázi infekce. Přesný mechanismus však není znám. Pro studium interakce proteinů VP2 a VP3 MPyV s membránou byly v minulosti vytvořeny expresní plasmidy, kódující mutované VP2 nebo VP3 fúzované s EGFP. Tyto proteiny mají delece v předpokládaných hydrofobních doménách. V této práci byla sledována lokalizace mutovaných proteinů v buňkách. Ukázalo se, že pro asociaci VP2 a VP3 s membránou je nejdůležitější hydrofobní doména 2, zatímco domény 1 a 3 jsou spíše postradatelné. Studium VP2 a VP3 pokračovalo dále analýzou charakteru jejich isoforem. Byly separovány isoformy obou proteinů o různé elektroforetické mobilitě pomocí SDS-PAGE. Následná analýza hmotnostní spektrometrií ukázala, že tyto isoformy se liší v důsledku deamidace na asparaginu společném oběma proteinům (pozice 253 pro VP2 a 137 pro VP3). Murine polyomavirus (MPyV) is a member of the Polyomaviridae family. Its capsid is composed of the major capsid protein, VP1, and the minor proteins, VP2 and VP3. The minor capsid proteins probably assure delivery of the viral genome through the endoplasmic reticulum membrane to the nucleus during early phase of infection. However, precise mechanism is not known. Expression plasmids encoding mutated VP2 or VP3 fused with EGFP have been constructed to study the interaction of VP2 and VP3 with membranes. The mutated proteins have deletions in the predicted hydrophobic domains. In this thesis, cell localisation of mutated proteins was followed. The study revealed that the hydrophobic domain 2 is the most important for association of VP2 and VP3 with membranes, while domains 1 and 3 are rather expendable. Further, nature of VP2 and VP3 isoforms has been studied. Isoforms with different electrophoretic mobility were separated on SDS-PAGE. Consequent mass spectrometry analysis showed that they differ in deamidation of asparagine, present at both minor proteins (position 253 of VP2 and 137 of VP3).
