5 2 SOUHRN Bioindikátory zánětu, oxidačního a nitračního stresu u potkana kmene Wistar po celotělovém a lokálním ozáření plic Úvod Radiační pneumonitida a fibróza limitují intenzitu radioterapie, zvyšují morbiditu a zhoršují kvalitu života nemocných s nádory plic a prsu, či Hodgkinovou nemocí. Radiační pneumonitida je akutní zánětlivá reakce, která ustupuje spontánně nebo po léčbě kortikosteroidy.Při jejím vzniku se významně uplatňují volné radikály. V závislosti na dávcevyvolávají lipoperoxidaci, poškození DNA, aktivaci proapoptotických dějů a změny aktivity enzymů. Na zánětlivé reakci iniciovanévolnými radikály se podílejí především epiteliální buňky a buňky zánětu, které uvolňují řadu cytokinů (TNF-α, IL-1, IL-6, TGF-β, aj.). Radiační fibróza plic se rozvíjí jako pozdní následek radioterapie (> 3 měsíce). Může vyústit do chronické respirační insuficience a způsobit smrt. Makroskopickým nálezem je proliferace fibroblastů, ukládání kolagenu a destrukce plicního parenchymu. Cíle práce a použité metody Předložená prácese zaměřila na změny v metabolické dráze arginin-oxid dusnatý vyvolané v dýchacích cestách a v jiných částech organismu ionizujícím gama zářením a to v časné fázi (do 72 h) a ve fázi radiační pneumonitidy (7 týdnů) po celotělové expozici i lokálním ozařování hrudníku potkana Wistar. 6 3 SUMMARY Bioindicators of inflammation, oxidative and nitrossative stress in Wistar rats after whole-body and lung irradiation Introduction Radiation pneumonitis and fibrosis limitthe intensity of radiotherapy,increase morbidity and worsen quality of life of patients with lung and breast cancer or Hodgkin's disease. Radiation pneumonitis is an acute inflammatory reaction that resolves either spontaneously or after treatment with corticosteroids. Free radicals play an important role in its etiology. They cause dose-dependent induction of lipoperoxidation, DNA damage, activation of proapoptotic events and changes in activity of enzymes. The inflammatory reaction initiated by free radicals affects primarily epithelial and inflammatory cells, which release a number of cytokines (TNF-α, IL-1, IL-6, TGF-β, etc.). Radiation fibrosis of lungdevelops as a late consequence of radiotherapy (> 3 months). It can lead to chronic respiratory failureand death.
