SCCmec, horizontal transfer, mobile genetic elements, MRSA, Staphylococcus spStafylokoky jsou běžnou součástí lidské flóry, ale jsou také nebezpečnými patogeny. Mezi stafylokoky je velmi rozšířená rezistence k meticilinu. Za rezistenci k meticilinu je odpovědný gen mecA uspořádaný do mec komplexu, který je součástí mobilního genetického elementu - stafylokokové chromozomové kazety SCCmec. SCCmec je rozsáhlý variabilní mobilní genetický element, je vždy tvořen třemi částmi, kterými jsou mec komplex, ccr komplex a J oblasti. Komplex mec obsahuje gen mecA a jeho regulační geny mecR1 a mecI. Komplex ccr kóduje geny pro rekombinázy, které jsou odpovědné za vystřižení a vložení SCCmec kazety. J oblasti jsou zbývající části SCCmec kazet, které obsahují další mobilní genetické elementy, ty mají buď přímý vliv na expresi genů pro rezistence k meticilinu, nebo nesou geny pro rezistence k jiným antimikrobiálním látkám. Celá kazeta nebo její součásti se mohou rozšiřovat horizontálním přenosem genů mezi stafylokoky stejného druhu i mezidruhově, i když mechanizmus přenosu je stále nejasný. V současnosti je popsáno celkem jedenáct typů SCCmec kazet. V této práci jsem shrnula poznatky o molekulárním složení SCCmec, horizontálním přenosu genů odpovědných za rezistenci k meticilinu a molekulární evoluci SCCmec. Staphylococci are common part of human flora but also they are a dangerous pathogen. Among staphylococci strains, methicillin resistance is widespread. The mecA gene, organized in mec complex, is responsible for methicillin resistance. The mec complex is part of mobile genetic element - staphylococcal chromosome cassette SCCmec. SCCmec is large variable mobile genetic element and it is always composed of three parts - mec complex, ccr complex and J regions. Complex mec consists of mecA gene and its regulatory genes mecR1 and mecI. Complex ccr encodes recombinase genes, they are responsible for excision and insertion of SCCmec. J regions are remaining parts of SCCmec, which include other mobile genetic elements that directly influence methicillin resistance genes expression or carry genes for resistance to other antimicrobial agents. SCCmec or its parts can be transferred by horizontal gene transfer between staphylococci both intraspecific and interspecific, although mechanism of its transfer is still unknown. Eleven types of SCCmec have been described so far.
