Modification of nanofibers, mesenchymal stem cells, polyvinylalcohol, electrospinning, tissue engineering of cartilageModifikace vláken za účelem funkcionalizace je aktuální trend v problematice tkáňového inženýrství. Polyvinylalkohol (PVA) je netoxický, biodegradabilní polymer vhodný k přípravě nanovláken metodou elektrostatického zvlákňování. Jeho nevýhodou je však velká rozpustnost ve vodě, která vede k okamžité degradaci vláken. Jako výhodné se pak ukazuje jeho chemické složení, kdy je možné modifikovat volné hydroxyskupiny PVA. Pro modifikace byla připravena PVA nanovlákna metodou elektrostatického zvlákňování z hladiny. Metodou acylace se podařilo navázat na povrch nanovláken linker tvořený z polyetylenglykolu (PEG) zakončený molekulou biotinu (PEG-b). Tato modifikace nenarušila vlákenný charakter PVA vrstvy. Vazbou linkeru PEG-b na PVA nanovlákna (PVA-PEG-b) došlo ke zvýšení stability vláken ve vodném prostředí a to až po dobu 41 dní, což je výrazné zvýšení oproti nemodifikované formě. Stabilita vláken ve vodném prostředí se zvyšuje s množstvím linkeru a metoda tedy umožňuje připravovat PVA nanovlákna o různé rozpustnosti. Výsledná modifikace se ukázala jako biokompatibilní pro mezenchymální kmenové buňky (MSC) i chondrocyty. Proliferace buněk na nosiči však byla nízká a kultivace obou typů buněk vykazovala postupný úbytek buněk, pravděpodobně z důvodu velké hydrofility nosiče. Z tohoto důvodu byly navrženy...
Modification of nanofibers is an actual trend in tissue engineering. Polyvinylacohol (PVA) is nontoxic and biodegradable polymer suitable for preparation of submicron fibers by electrospinning. Main disadvantage of PVA fibers is rapid degradation in aqueous environment. On the other hand surface of fibers contains free hydroxyl group that could be chemically modified. In recent work, chemical modification of PVA nanofibers prepared by needleless electrospinning was investigated. Polyethylenglykol (PEG) linker was introduced to the fiber surface by acylation (PVA-PEG) and further modified by biotin (PVA-PEG-b) as a function agent. Process of chemical modification does not affected fibrous morphology of samples. Interestingly, linkage of PEG-b linker promoted stability of PVA in aqueous environment. PVA-PEG-b sample was stable for 41 days. Stability of samples was strongly dependent on amount of introduced PEG-b linker, thus proposed method of modification allows to prepare nanofibers of different solubility. Additionally, biocompatibility of chemically modified nanofibers with both mesenchymal stem cells (MSC) and chondrocytes was determined.
