Katecholaminová kardiotoxicita je známa již dlouhou dobu, avšak její patogeneze stále není dostatečně objasněna. Ačkoliv se tradičně přisuzuje nad- měrné β-adrenergní stimulaci, existuje hypotéza, že do katecholaminy- navozeného poškození srdečních buněk jsou zahrnuty oxidační produkty kate- cholaminů, které mohou vznikat díky spontánní oxidaci katecholaminů za ae- robních podmínek. Spontánní oxidace katecholaminů je sledem redoxních re- akcí vedoucích k tvorbě řady reaktivních meziproduktů, jako jsou o-semichino- ny a o-chinony včetně aminochromů, které podléhají redoxní cyklizační reakci generující reaktivní formy kyslíku (ROS). Vytvořené ROS i aminochromy jsou pro buňky toxické. Navíc může být tato autooxidace katalyzována přechodný- mi kovy, z čehož vyplývá, že volné vnitrobuněčné ionty železa (Fe) - nejhoj- nějšího přechodného kovu v organizmu - mohou podporovat nejen Haber- Weissovu reakci ROS, ale také katalyzovat přeměnu katecholaminů na ami- nochromy, a tím zhoršovat...

Catecholamine cardiotoxicity is known for a long time, but its patho- genesis is still incompletely understood. Although traditionally attributed to excessive β-adrenergic stimulation, there is a hypothesis that the catecholamine- induced cardiac cell damage includes involvement of oxidation products of catecholamines, which may arise due to spontaneous oxidation of catechola- mines under aerobic conditions. Spontaneous oxidation of catecholamines is a sequence of redox reactions leading to the formation of numerous reactive in- termediates, such as o-semiquinones and o-quinones, including aminochromes that are subject to redox cyclization generating reactive oxygen species (ROS). The generated ROS and aminochromes are toxic for cells.
