Optimalizace chromatografických podmínek pro HPLC stanovení biologicky aktivních látek Diplomová práce Eva Kotíková Univerzita Karlova v Praze, Farmaceutická fakulta v Hradci Králové, Katedra farmaceutické chemie a kontroly léčiv Adenosintrifosfát (ATP), adenosindifosfát (ADP), adenosinmonofosfát (AMP), kreatin (C) a fosfokreatin (CP) jsou polární látky s velkým významem nejen v lidském organismu. V této práci se podařilo vyvinout HPLC/UV metodu, která umožňuje separaci všech těchto látek za izokratické eluce. Jako vhodná stacionární fáze byla vybrána zirkoniová kolona, jejíž hlavní předností oproti klasickým silikagelovým kolonám je stabilita v celé škále pH a při teplotách až do 200žC. Nejlepší separace tak bylo dosaženo na koloně ZirChrom® - PHASE, s použitím dvousložkové mobilní fáze s průtokem 1 ml/min - složka A: 10mM hydrogenfosforečnanový pufr, pH 7 a složka B: 100% acetonitril. Detekce byla prováděna pomocí UV detektoru při vlnových délkách 254 nm pro ATP, ADP, AMP, cAMP a při 215 nm pro C a CP. Výsledné retenční časy při těchto podmínkách byly: cAMP = 2,1 min, C = 3,7 min, AMP = 4,7 min, CP = 6,4 min, ADP = 10,7 min, ATP = 27,9 min. In this work HPLC / UV method which allows the separation of these substances in isocratic elution was developed. As a suitable stationary phase zirconia column was chosen. The main advantage in comparison with the traditional silica columns is stability in the whole range of pH as well as at temperatures up to 200 žC. The best separation was achieved on a ZirChrom® - PHASE column by using a binary mobile phase with flow rate 1 ml/min - Part A: 10 mM hydrogenphosphate buffer, pH 7 and Part B: 100% acetonitrile. The detection was performed by using a UV detector at wavelengths of 254 nm for ATP, ADP, AMP, cAMP and 215 nm for C and CP.
