Nejen samotná hypoxie mozku, ale i následná reperfúze vyvolává závažné změny vnitřního prostředí, které ve svých důsledcích mění funkce a strukturu nervových i gliových buněk. Z experimentálních nálezů vyplývá, že existuje řada iontů (např. Mg2+), které mohou tyto negativní změny v mozku zmírnit. Za použití histochemické metody (NADPH-d diaforáza barvení) byl zjišťován vliv magnézia na denzitu NADPH-d pozitivních neuronů v hippocampu potkanů vystavených dlouhodobé opakované hypoxii v období rané ontogeneze (12, 25 a 35 denní zvířata). NADPH-diaforáza (NADPH-d) je enzym kolokalizovaný s nitric oxid syntázou (NOS), která katalyzuje syntézu oxidu dusnatého (NO). Výsledky ukázaly, že dlouhodobá intermitentní hypobarická hypoxie zvyšuje denzitu NADPH- pozitivních neuronů ve všech sledovaných oblastech u 12 denních zvířat, u 25 denních zvířat pouze v hilu gyrus dentatus a u 35 denních zvířat v CA1, CA3 oblastech hippocampu a ve ventrálním listu gyrus dentatus. Naopak snížení denzity nitrergních neuronů následkem hypoxie bylo zjištěno u 25 denních mláďat v obou listech gyrus dentatus a u 35 denních zvířat jen ve ventrálním listu a v hilu gyrus dentatus. Premedikace magnéziem u zvířat vystavených hypoxii způsobila snížení denzity nitrergních neuronů ve všech sledovaných oblastech hippocampu a to ve všech věkových...
Hypoxia of the brain as well as the subsequent reperfusion can seriously alter the tissue microenvironment and result of the functional and structural changes of nerve and glial cells. Results of experimental studies show that some ions (e.g. Mg2+) can interfere with the brain development. To answer the question whether magnesium can modulate changes of neuronal circuits induced by hypoxia and reperfusion effect of magnesium administration on the density of nitrergic neurons (NO synthesising neurons) in the rats exposed to repeated hypoxia during the postnatal ontogeny (12, 25, and 35-day-old) was studied. NO synthesising neurons were identified according to the presence of NADPH-diaphorase (NADPH-d), the enzyme co-localized with NO synthase. Results have shown that the long-lasting intermittent hypobaric hypoxia brings about the increase of the density of NADPH-diaphorase positive neurons in all studied regions of the hippocampus in 12-day-old animals, in 25-day-old only in the hilus of the dentate gyrus, and in 35-day-old in CA1, CA3 hippocampal regions and in the ventral blade of the dentate gyrus.
